HPLC法检测不同温度和时间对蜂王浆中癸烯酸的影响

【摘要】 采用高效液相色谱法,对不同贮藏温度和时间的蜂王浆中10-羟基-2-癸烯酸(10-HDA)含量进行测定。样品采用95%乙醇溶解,采用C18柱,流动相为v(甲醇):v(0.1%甲酸)=55:45,流速0.5mL/min,检测波长210 nm,采用外标定量法进行测定,10-HDA在质量浓度为50~400 mg/L范围内呈良好线性关系,相关系数R=0.9996。研究表明:该方法简便、快速、准确、可靠,在不同温度和时间下蜂王浆中癸烯酸含量较稳定。

【Abstract】 High performance liquid chromatography was used for the determination of 10-hydroxy-2-decenoic acid in royal jelly at different temperature and time. Sample was extracted by 95% ethanol, chromatographic separation was performed using C 18 column with methanol and 0.1% formic acid(v/ v, 55:45) as mobile phase at a fl ow rate of 0.5 mL/min. The UV detection wavelength was fi xed at 210 nm, external calibration method was used in this experiment the linearity was obtaied over the range of 50~400 mg/L(R=0.9996). The results showed that this method is simple, quick, specifi c and accurate, and 10-hydroxy-2-decenoic acid was stable at different temperature and time.
