2种膨松剂对小鼠遗传毒性的比较研究
Comparative study of two swelling agents in genotoxicity of mice
【摘要】 目的:观察硫酸铝钾和碳酸氢铵对小鼠骨髓细胞微核率和精子畸形率的影响,了解是否具有遗传毒性。方法:以环磷酰胺(45 mg/kg)为阳性对照,蒸馏水为阴性对照,不同浓度的膨松剂(正常添加剂量、中间剂量和LD50剂量)为试验组,经腹腔注射,连续给药4 d,观察小鼠骨髓嗜多染红细胞微核率和精子畸形率。结果表明:硫酸铝钾各剂量组诱导小鼠产生的微核率明显高于阴性对照组(P<0.05或P<0.01),但低于环磷酰胺阳性对照组;碳酸氢铵各剂量组诱导小鼠产生的微核率与阴性对照相比无显著性差异。2种膨松剂各剂量组均能诱发小鼠精子产生畸形,随着浓度的增大,小鼠精子畸形率增大。除碳酸氢铵正常添加剂量组外,其余各组精子畸形率与阴性对照组相比,差异显著(P<0.05)或极显著(P<0.01)。结论:硫酸铝钾具有一定的遗传毒性,碳酸氢铵正常添加剂量没有遗传毒性。
【Abstract】 Objective: To investigate effects of aluminum potassium sulfate and ammonium bicarbonate on micronucleus rates of bone marrow cells and the rate of sperm abnormality in mice to identify whether two swelling agents have genetic toxicity. Methods: The micronucleus rate of bone marrow polychromatic erythrocytes and the rate of sperm abnormality were determined by using intraperitoneal injection with different concentrations of the swelling agents(normal amount of additive, intermediate dose and LD 50 dose) at the same time cyclophosphamide(45 mg/kg) as the positive contrast and water as the negative contrast for 4 days. Results: the micronucleus rates of aluminium potassium sulfate in every groups are obviously higher than the rate of the negative control group(P<0.05 or P<0.051), but lower than the rate of the positive control group(CP). The rates of ammonium bicarbonate groups are no significant difference with the rate of the negative control group. Each dose group of two swelling agents can induce sperm deformity in mice. Their malformation rate increases with the increase of the concentration. Except the normal amount of additive in ammonium bicarbonate, the rate of sperm abnormality are significant difference with the rate of negative control group in other groups. Conclusion: Aluminium potassium sulfate has certain genetic toxicity. Ammonium bicarbonate has no genetic toxicity in normal amount of additive.
