毛细管电泳分离检测茶叶中5种多酚类化合物
Separation and Determination of Five Kinds of Polyphenolic Compounds in Tea by Capillary Electrophoresis
【摘要】 利用高效毛细管区带电泳,建立一种同时分离测定茶叶中儿茶素、表儿茶素、槲皮素、山奈酚和杨梅素的方法。在含有10 mmol/L Na2B4O7、5 mmol/Lβ-环糊精及8%(V/V)乙腈的运行缓冲溶液(pH 9.11)中,采用20 kV的分离电压、25℃的毛细管柱温和200 nm的检测波长,儿茶素、表儿茶素、槲皮素、山奈酚和杨梅素可以在14 min实现有效分离与检测,将方法用于不同茶叶样品中这5个组分的测定,相对标准偏差在4.0%以内,回收率为95.4%~104.6%。本方法简单、快速、线性范围较宽、重复性好,可用于茶叶中这5个组分的测定。
【Abstract】 Objective: To develop a method for simultaneous separation and determination of catechin, epicatechin, quercitin, kaempferol and myricetin in tea. Methods: High performance capillary zone electrophoresis was used. Results: Catechin, epicatechin, quercitin, kaempferol and myricetin could be effectively separated and determined within 14 min in a running buffer solution(pH 9.11) containing 10 mmol/L Na2B4O7, 5 mmol/L β-CD and 8%(V/V) acetonitrile with the applied voltage of 20 kV, capillary temperature of 25 ℃ and detector wavelength of 200 nm. The relative standard deviation and recovery of the proposed method for these five compounds in tea were below 4.0% and in the range of 95%-105.6%, respectively. Conclusion: This method possesses the advantages of simple operation, rapid determination, high reproducibility and wide detection range. It can be used for the determination of catechin, epicatechin, quercitin, kaempferol and myricetin in tea.
