益眠达片对失眠大鼠下丘脑单胺类神经递质的影响

【摘要】 目的观察益眠达片对失眠大鼠下丘脑单胺类神经递质含量的影响,探讨其治疗失眠的作用机制。方法将大鼠随机分为空白对照组、模型对照组、益眠达片组,艾司唑仑组,每组10只。除空白对照组外,其余各组大鼠均用对氯苯丙氨酸建立大鼠失眠模型。益眠达片组和艾司唑仑组分别给予益眠达片和艾司唑仑混悬液灌胃,其余2组给予生理盐水灌胃。6 d后,采用荧光分光光度计测定下丘脑内去甲肾上腺素(NE)及5-羟色氨(5-HT)、5-羟吲哚醋酸(5-HTAA)含量。结果与模型对照组比较,益眠达片组可以降低大鼠下丘脑内NE含量、升高下丘脑内5-HT、5-HTAA含量(P<0.01)。结论益眠达片改善失眠大鼠唾眠的机制与其影响大鼠下丘脑内单胺类神经递质含量有关。

【Abstract】 Objective The influence of Yimianda tablets on the contents of monoamines neurotransmitters in Hypothalamus of insomnia rats,to explore the mechanism of curative effect of Yimianda tablets. Me thods 40 Wistar rats were randomized into the blank control group,model group,Yimianda tablets group,and Estazolam group,each group had 10 rats. The experimental model of insomnia rats was induced by PCPA. The rats of Yimianda tablets group give Yimianda Suspension gavaged,The rats of Estazolam group give Estazolam Suspension gavaged. The rats of blank control group and model group give NS gavaged. After 6 days,used fluorescence spectrophotometer determined the contents of NE、5-HT、5-HTAA in the hypothalamus of insomnia rats. Re sults Compare with the model group’s rats,the Yimianda tablets group’s rats’ the contents of NE was reduced and the contents of 5-HT、5-HTAA was increased in the hypothalamus( P < 0.01). Conclusion The mechanism of Yimianda tablets’ improve insomnia related to influence the contents of monoamines neurotransmitters in hypothalamus of insomnia rats.
