白藜芦醇对内皮细胞的保护作用及机制研究
Research on Protective Effects and Mechanism of Resveratrol on Endothelial Cells
【摘要】 目的:研究白藜芦醇(Res)对动脉粥样硬化(AS)内皮细胞损伤的保护作用及机制。方法:应用溶血性磷脂酰胆碱(LPC)诱导的内皮细胞损伤模型,用双染流式细胞技术测定细胞凋亡率,TUNEL染色实验测定细胞凋亡数,MTT比色实验测定细胞活力,自动生化仪测定细胞培养基乳酸脱氢酶(LDH)水平,ELISA法测定TNF-α含量,RT-PCR测定CGRP mRNA水平。结果:20μmol·L-1浓度Res可有效地减弱由LPC诱导的细胞活力下降、培养基LDH水平升高及TNF-α含量升高(P<0.01),Res可促进CGRP mRNA表达。结论:Res具有抗AS内皮损伤作用,其机制可能与通过激活VR1促进CGRP的合成和释放有关。
【Abstract】 This article was aimed to study the protective effects and mechanism of resveratrol(Res) on endothelial injury induced by atherosclerosis(AS). Lysophosphatidylcholine(LPC) was applied to induce the model of injured endothelial cells. Flow cytometer was used to detect the rate of cell apoptosis. TUNEL staining was used to detect the cell apoptosis. MTT colorimetric test was used to detect the cell viability. Automatic biochemistry analyzer was used to measure the LDH level. ELISA was used to detect TNF-α level. RT-PCR was used to determine the mRNA levels of calcitonin gene-related peptide(CGRP). The results showed that Res with the concentration of20 μmol/L can effectively slow down the degree of endothelial cell injury induced by LPC which caused the cell disability, high LDH level and high TNF-α level(P < 0.01). Res showed effects on mRNA expression of CGRP.It was concluded that Res can protect against endothelial injury induced by AS, the mechanism of which may be associated with accelerating CGRP synthesis and release by activating VR1.
