【摘要】 目的:探讨caspase 8在热疗降低胶质瘤侵袭性过程中的作用。方法:利用Transwell构建胶质瘤侵袭模型,随机分为热疗对照组以及热疗30、60、120、180 min和240 min组。采用流式细胞术检测胶质瘤细胞凋亡水平,免疫印迹法检测胶质瘤细胞caspase 8的表达水平。结晶紫染色法测定胶质瘤侵袭性变化。结果:(1)热疗各组胶质瘤细胞凋亡率均明显高于热疗对照组(P<0.05);(2)各热疗组胶质瘤细胞的caspase 8表达均高于热疗对照组,有统计学差异(P<0.05);(3)各热疗组胶质瘤细胞的侵袭性均较对照组降低(P<0.05)。结论:热疗后胶质瘤细胞侵袭性降低,这可能与caspase 8表达增加导致胶质瘤细胞凋亡有关。更多还原

【Abstract】 Objective: To probe the role of caspase 8 in the process of hyperthermia reducing glioma invasiveness. Methods: Transwell was used to make glioma invasion models which were randomly divided into control group and hyperthermia groups( 30,60,120,180 and 240 min). Flow cytometry( FCM) was employed to test the quantification of the apoptotic glioma cells,Western-Blot was used to detect the expression of caspase 8,and crystal violet staining was used to detect the change of glioma cells invasiveness. Results:( 1) The apoptotic cell rate of hyperthermia groups was significantly higher than that of the control group( P < 0. 05).( 2) The caspase 8 expression in hyperthermia groups was higher than in the control group,and the difference was statistically significant( P < 0. 05).( 3) The glioma cells invasiveness of hyperthermia groups was lower than that of control group( P < 0. 05). Conclusion: Glioma cell invasion is reduced after hyperthermia,which may be related to the glioma cells apoptosis caused by the increased expression of caspase 8.更多还原
