NF-κB p50、NF-κB p65在甲状腺癌中的表达及意义
Expression and significance of NF-κB p50 and NF-κB p65 in thyroid cancer
【摘要】 目的探讨NF-κB p50、NF-κB p65蛋白在甲状腺癌、甲状腺腺瘤、结节性甲状腺肿、正常甲状腺组织中的表达及二者与甲状腺癌的浸润、转移的关系。方法采用免疫组织化学染色法检测NF-κB p50和NF-κB p65蛋白在甲状腺癌组(60例乳头状癌、20例滤泡癌和20例髓样癌)及非甲状腺癌组(30例健康者、30例甲状腺腺瘤、30例结节性甲状腺肿)的甲状腺组织的表达,并检测NF-κB p50和NF-κB p65蛋白的表达的相关性。结果 NF-κB p50蛋白在甲状腺癌、甲状腺腺瘤、结节性甲状腺肿及健康者甲状腺组织中的阳性率分别为73.00%、30.00%、33.33%及6.67%。NF-κB p50蛋白在甲状腺非癌组织的阳性率明显低于癌组织。NF-κB p65蛋白在甲状腺癌、甲状腺腺瘤、结节性甲状腺肿及健康者甲状腺组织中的阳性率分别为74.00%、36.67%、40.00%及10.00%。NF-κB p65蛋白在甲状腺非癌组织的阳性率明显低于癌组织。NF-κB p50蛋白在肿瘤直径≥1.0 cm组及<1.0 cm组的阳性率分别为79.41%和56.25%,差异均有统计学意义(P<0.05);NF-κB p65蛋白在肿瘤直径≥1.0 cm组及<1.0 cm组阳性率分别为94.12%和81.25%,差异均有统计学意义(P<0.05)。NF-κB p50蛋白和NF-κB p65蛋白在淋巴结转移组的阳性率分别为85.19%和87.04%;在无淋巴结转移组的阳性率分别为56.52%%和69.57%。差异均有统计学意义(P<0.05)。NF-κB p50蛋白和NF-κB p65蛋白的表达呈正相关关系(r=0.712,P<0.05)。结论 NF-κB p50蛋白和NF-κB p65蛋白参与甲状腺癌的表达,并于甲状腺癌的发生、发展过程中发挥着重要的协同作用。
【Abstract】 Objective To investigate the expression of NF-κB p50 and NF-κB p65 protein in thyroid cancer,thyroid adenoma,nodular goiter and normal thyroid tissues,and the role of NF-κB p50,NF-κB p65 protein in infiltration and metastasis of thyroid cancer. Methods Immunohistochemical staining was used to detect the expression NF-κB p50 and NF-κB p65 protein in thyroid tissue in thyroid cancer group(60 cases of papillary carcinoma,20 cases of follicular carcinoma,20 cases of medullary carcinoma) and non-cancerous group(30 cases of healthy thyroid,30 cases of thyroid adenoma,30 cases of nodular goiter),and correlation of expression between NF-κB p50 and NF-κB p65 protein. Results In thyroid tissues of 100 patients with thyroid cancer,the positive rate of NF-κB p50 protein expression was 73. 00%; in thyroid tissues of 30 patients with thyroid adenoma and 30 patients with nodular goiter,the positive rate were 30. 00% and 33. 33% respectively; in thyroid tissues of 30 healthy people,the positive rate was 6. 67%,the positive rate of NF-κB p50 protein expression in thyroid non-cancerous tissues was significantly lower than that of thyroid cancer tissue. In thyroid tissues of 100 patients with thyroid cancer,the positive rate of NF-κB p65 protein expression was 74. 00%; in thyroid tissues of 30 patients with thyroid adenoma and 30 patients with nodular goiter,the positive rate were 36. 67% and 40. 00% respectively; in thyroid tissues of 30 healthy people,the positive rate was 10. 00%. The positive rate of NF-κB p65 protein expression in non-cancerous tissues was significantly lower than that of thyroid cancer tissue. In tumor diameter≥1. 0 cm group and tumor diameter < 1. 0 cm group,the positive rate of NF-κB p50 protein expression were 79. 41% and 56. 25%respectively,and the difference was statistically significant(P < 0. 05); in tumor diameter≥1. 0 cm group and tumor diameter < 1. 0 cm group,the positive rate of NF-κB p65 protein expression was 94. 12% and 81. 25% respectively,and the difference was statistically significant(P < 0. 05). The positive rate of NF-κB p50 protein and NF-κB p65 protein in lymph node metastasis group were 85. 19% and 87. 04%,respectively; the positive rate in the group without lymph node metastasis were 56. 52% % and 69. 57%,respectively. The differences were statistically significant(P < 0. 05). The correlation of expression between NF-κB p50 protein and p NF-κB p65 positive was positive(r = 0. 712,P < 0. 05). Conclusion NF-κB p50 protein and NF-κB p65 protein are involved in the expression of thyroid cancer,and play an important synergetic role in the occurrence and development of thyroid cancer.
