SDF-1在牵张成骨中的表达及作用机制研究

【摘要】 目的探讨基质细胞衍生因子-1(stromal cell-derived factor-1,SDF-1)在大鼠下颌骨中牵张成骨(distraction osteogenesis,DO)区的表达情况及其作用机制。方法 40只SD大鼠随机分为8组,每组各5只,全部将右侧下颌骨表面皮肤切开,放置下颌骨牵张器,调整塑形并固定。而后将大鼠左侧下颌骨骨折行钛板内固定手术以作对照。随后分别于不同的时间点(0、3、6、9、12、15、18、21 d)将大鼠麻醉处死,解剖大鼠双侧下颌骨后,小心刮取右侧牵张间隙骨痂及左侧骨折处骨痂进行免疫组织化学检测和酶联免疫吸附测定。结果 40只大鼠伤口均愈合正常,手术耐受性良好。免疫组化染色结果显示:SDF-1在成骨细胞和微血管周围表达较多,且在DO的新生骨痂中的表达较骨折处骨痂中的表达明显升高。酶联免疫吸附检测结果显示:无论在DO区或是骨折区,SDF-1术后3 d的表达量增加了3倍,术后1周后,SDF-1在骨折区的表达情况呈逐渐下降趋势,在DO区的表达在进入牵张期内其再次上升,高达原有水平的5倍,牵张结束后缓缓下降,SDF-1在2区的表达情况比较分别在9、12、15 d天有统计学差异(P<0.05)。结论 SDF-1在牵张成骨区和骨折区的表达差异明显,为日后研究SDF-1在牵张成骨中的表达及作用机制奠定了理论基础。

【Abstract】 Objective To explore the expression of stromal cell-derived factor-1 in rat mandibular bone area in distraction osteogenesis and its mechanism of action. Methods 40 SD rats were randomly divided into eight groups,each had five rats,all the surface of skin incision on the right side of mandible,mandibular distractor placement,adjust shaping and fixed. The lower left side of the rat mandibular fractures after internal fixation with a titanium plate as controls. Subsequently,rats were sacrificed at different time points(0,3,6,9,12,15,18,21 d),dissected rat bilateral mandible,carefully scraping the callus from the right side of the distraction gap and left side of the fracture zone for chemical tests and enzyme-linked immunosorbent assay immunohistochemistry. Results 40 rats were all in normal wound healing with good operation tolerance; Immunohistochemical staining showed that,the expression of SDF-1 in osteogenic cells and capillaries around were increased,and its expression in DO freshmen zone was more than fracture zone; ELISA test results showed: the expression of SDF-1 at 3 days after operation were increased triple both in DO zone and fracture zone.And one week after surgery,SDF-1 expression in the fracture zone was decreased,and in DO zone rising again before entering the distraction period,up to five times,the differences of SDF-1 expression between two zone at 9,12,15 d were all significant(P < 0. 05). Conclusion SDF-1 expression levels were different in distraction bone area and fracture zones,which can lay the foundation for future research of SDF-1 expression in distraction and its mechanism.
