复发性流产患者蜕膜组织NK细胞受体的表达

【摘要】 为探讨子宫蜕膜组织NK细胞受体的表达与复发性流产发病之间的关系,采用实时荧光定量RT-PCR和免疫组织化学法检测复发性难免流产及正常早孕蜕膜组织的NK细胞受体KIR2DL4、ILT-2、ILT-4 mRNA及其蛋白的表达水平。研究结果显示:难免流产组KIR2DL4、ILT-2、ILT-4 mRNA水平明显低于正常妊娠组,免疫组化的结果也显示难免流产组KIR2DL4、ILT-2的分布密度低于正常妊娠组。该结果提示:NK细胞受体的表达水平与难免流产有一定关联。推测复发性流产患者KIR2DL4表达下调,可能会影响早孕期蜕膜化的关键进程,进而对妊娠结局产生不良影响;而ILT-2、ILT-4表达下调,可能使得免疫细胞活化阈值下降,导致局部效应细胞功能紊乱,最终发生流产。

【Abstract】 To study on the relationship between expression of NK cell receptors on placenta decidual tissue in recurrent spontaneous abortion(RSA)and the pathogenensis of RSA,mRNA expression of NK cell receptors KIR2DL4,IL T-2 and IL T-4 on placenta decidual tissue from RSA women and normal early pregnant women was examined by real-time RT-PCR,and the protein expression of KIR2DL4,IL T-2 and IL T-4 were examined by immunohistochemistry with monoclonal antibodies mAb#33,HP-F1 and Anti-Human IL T-4 which are respectively specific for the above mentioned receptors.The results of real-time RTPCR and immunohistochemistry showed that the levels of KIR2DL4,IL T-2,IL T-4 on placenta decidual tissue from RSA were much lower than those from the normal controls.These results indicate that there may be some association between the low level gene expression of KIR2DL4,IL T-2,IL T-4 and RSA.The potential consequences of low expression of these natural killer receptors might change the secretion pattern of the infiltrating immune ceils and decreasethe activation threshold of NK cells and cytotoxic T lymphocytes,and finally result in the failure of pregnancy.
