睡眠剥夺对大鼠颞下颌关节髁突组织p38信号通路相关蛋白表达的影响

【摘要】 目的探讨睡眠剥夺对大鼠颞下颌关节(TMJ)的破坏作用及对p38信号通路相关蛋白表达的影响。方法将40只Wistar雄性大鼠随机分为SD组和对照组。SD组采用改良多平台法建立睡眠剥夺模型。于制模4 d后处死两组大鼠,HE染色观察TMJ髁突组织病理学变化,免疫组化SP法检测TMJ髁突组织p38信号通路相关蛋白MKK6、p38表达。结果 SD组TMJ髁突表面可见部分胶原纤维水肿、松解,出现炎症反应;SD组MKK6、p38阳性细胞率均高于对照组;MKK6、p38蛋白表达阳性率均高于对照组(P<0.05)。结论大鼠睡眠剥夺时TMJ出现病理性改变,p38信号通路可能参与了TMJ关节破坏的病理过程。

【Abstract】 Objective To observe the structural changes and expression of p38 pathway related proteins in the rats’ temporomandibular joints( TMJ) after sleep deprivation. Methods Forty male Wistar rats were randomly divided into two groups: the sleep deprivation( SD) group and the control group. The sleep deprivation models were established by the modified multiple platform method. Rats of each group were sacrificed after 4 days sleep deprivation,and stained with HE to observe the morphological changes,and the expression of MKK6 and p38 protein in the TMJ condyle was detected by immunohistochemical SP method. Results In the SD group,edema and releases of the collagen fiber in the condylar surface were observed,and the inflammatory reaction appeared. Immunohistochemistry SP method indicated that the phosthorylated MKK6 and phosthorylated p38 positive cells and protein expression in SD group were more than those of the control group( P <0. 05). Conclusion The pathological changes of TMJ appears in rats undergoing sleep deprivation,and p38 pathway may take part in the TMJ pathological process.
