【摘要】 目的:研究糖肝消颗粒对糖尿病大鼠肝脏功能及形态的防治作用及其作用机制。方法:随机将40只健康Wistar大鼠分为4组,正常对照组(N)10只,糖尿病模型组(DM)10只,糖肝消组(TX)10只,二甲双胍组(DT)10只。大鼠腹腔注射链脲佐菌素建立糖尿病大鼠模型后,予以高脂饲料喂养8周,随机取3只大鼠进行形态学观察,符合NAFLD病理学改变,证实DM合并NAFLD模型成功。连续给予糖肝消及二甲双胍8周后观察大鼠体重、肝指数、血脂(TG、TCH)及肝功(ALT、AST)、空腹胰岛素(FINS)、胰岛素抵抗指数(IR)、空腹血糖(FPG),并通过光镜观察肝形态学变化。结果:与模型组比较,糖肝消可明显降低大鼠的体重、肝指数(P<0.05),与二甲双胍组比较无明显差异(P>0.05);明显降低大鼠空腹胰岛素(FINS)、胰岛素抵抗指数(IR)、空腹血糖(FPG)(P<0.01),疗效优于二甲双胍(P<0.01);可明显降低血脂(TG、TCH)(P<0.01)及肝功(ALT、AST)(P<0.05),疗效优于二甲双胍(P<0.01,P<0.05)。光镜观察表明,糖肝消对DM合并NAFLD大鼠肝脏有保护作用,TX组肝组织弥漫性肝细胞脂肪变较模型组程度明显减轻,肝脂肪变轻微。结论:糖肝消颗粒可以有效改善DM合并NAFLD大鼠IR,并辅助降糖,保护肝功能,修复受损的肝脏形态,为中医临床防治NAFLD的研究提供支持。更多还原

【Abstract】 Objective: To Study the effect and mechanism of Tangganxiao Granule on liver shape of diabetic rats. Methods:40 health Wistar rats randomly divided into 4 groups,normal controlled group( N),diabetic model group( DM),Tangganxiao Granule group( TX) 10,metformin group( DT),each group was 10 rats. Urea with intraperitoneal injection of rats chain rhzomorph diabetic rats model was established,high-fat feed 8 weeks,randomly took three morphological observation in rats,according with NAFLD pathology change,confirmed the DM merger NAFLD model success. Having been given Tangganxiao Granule and metformin for eight consecutive weeks and observing rat body weight,liver index,blood lipid( TG,TCH) and hepatic function( ALT,AST),fasting insulin( FINS),insulin resistance index( IR),fasting plasma glucose( FPG),and observing liver morphological changes. Results: Compared with model group,Tangganxiao Granule can obviously reduce the weight and liver index of rats( P < 0. 05),compared with metformin group,there was no significant difference( P >0. 05);rats significantly reduced fasting insulin( FINS),insulin resistance index( IR),fasting plasma glucose( FPG)( P < 0. 01),the curative effect was superior to metformin( P < 0. 01); could obviously reduce blood lipid( TG,TCH)( P < 0. 01) and hepatic function( ALT,AST)( P < 0. 05),the curative effect was superior to metformin( P < 0. 01,P < 0. 05). It showed that the Tangganxiao Granule on DM combined NAFLD rat liver protective,TX group of liver tissue diffuse hepatocellular fatty was changed,compared with model group was significantly reduced,liver fatty changed slightly. Conclusion: Tangganxiao Granule can effectively improve IR of DM rats combining NAFLD,and auxiliary hypoglycemic,protect the liver function,repair the damaged liver morphology,provide support for the research of TCM clinical prevention and treatment of NAFLD.更多还原
