【摘要】 目的:通过研究茵栀黄注射液对TNF-α作用下大鼠原代肝细胞Mrp2表达的影响,分析茵栀黄注射液的利胆退黄作用机制。方法:三明治法培养大鼠原代肝细胞。以茵栀黄注射液和/或TNF-α处理大鼠原代肝细胞。以定量PCR和Western-blot检测大鼠原代肝细胞Mrp2 mRNA及蛋白的表达。结果:对照组肝细胞Mrp2 mRNA表达高于TNF组(0.568±0.056,P<0.05),低于茵栀黄组(1.591±0.107,P<0.05);茵栀黄加TNF组肝细胞Mrp2 mRNA(1.079±0.087)表达高于TNF组(P<0.05)。对照组肝细胞Mrp2蛋白(0.768±0.070)表达高于TNF组(0.119±0.031,P<0.05),低于茵栀黄组(1.895±0.129,P<0.05);茵栀黄+TNF组肝细胞Mrp2蛋白表达(1.118±0.091)高于TNF组(P<0.05)。结论:茵栀黄注射液能改善TNF-α所导致的大鼠原代肝细胞Mrp2表达下降。更多还原

【Abstract】 Objective: To investigate the choleretic and jaundice-relieving mechanism of Yinzhihuang injection,the influence of Yinzhihuang injection on the Mrp2 expression in rat primary hepatocytes under the action of TNF-α. Methods: Primary rat hepatocytes were cultured in sandwich configuration. Primary rat hepatocytes were treated with Yinzhihuang injection and / or TNF-α. The expressions of Mrp2 mRNA / protein were detected with quantitative PCR and Western-blot. Results: The Mrp2 mRNA expression level of hepatocytes in controlled group was higher than that in TNF group( 0. 568±0. 056,P < 0. 05),lower than that in Yinzhihuang group( 1. 591±0. 107,P < 0. 05). The Mrp2 mRNA expression level of hepatocytes in Yinzhihuang adding TNF group( 1. 079±0. 087) was higher than that in TNF group( P < 0. 05).
