四川地区北京型结核分枝杆菌的MIRU-VNTR基因分型
Genotyping of Beijing family of mycobacterium tuberculosis isolates from sichuan with MIRU-VNTR
【摘要】 用寡核苷酸多态性基因分型(Spoligotyping)对2008年1月至2010年10月间采集的211株结核分枝杆菌临床分离株进行基因分型,然后对北京型结核分枝杆菌VNTR位点的多态性进行检测.选用的12个VNTR位点存在较高的基因多态性,该12位点组合分辨能力(HGI=0.99985)高于标准的VNTR-12位点组合(HGI=0.99546),其中多态性较高的7位点组合HGI指数为0.99977.因此,7位点组合可以作为四川地区北京家族分枝杆菌的一线分型方法.
【Abstract】 Clinical M.tuberculosis isolates from Jan 2008 to Oct 2010 were genotyped by Spoligotyping,and Beijing family was further analyzed for the polymorphism of the twelve tandem repeat loci.Analysis based on MIRU-VNTR for 162 Beijing isolates suggested that the selected 12 loci show a higher HGI value compared with the original 12 loci method.Using 7-locus VNTR of high polymorphism,the HGI value(0.9998)was obtained.It is suggested that the method of 7-locus can be used as the first-line molecular typing tool for Beijing family isolates of M.tuberculosis.
