TE、LOXL-1和Elastase在腹股沟疝患者腹横筋膜中的表达及意义

【摘要】 目的探讨弹性蛋白原(TE)、类赖氨酰氧化酶-1(LOXL-1)和弹性蛋白酶(Elastase)在腹股沟疝患者腹横筋膜中的表达及意义。方法选择佛山市第一人民医院行择期疝修补术的腹股沟疝患者60例,分为腹股沟直疝组(A组)和腹股沟斜疝组(B组),每组30例;另外选取同期行腹部手术的30例患者(非疝、非炎性疾病)作为对照组(C组),分别采用免疫组化和Western blot方法检测各组腹横筋膜中TE、LOXL-1和Elastase的表达。结果三组患者一般资料比较差异无统计学意义(P>0.05);免疫组化和Western blot方法均显示TE和LOXL-1在腹横筋膜中的表达为:C组>B组>A组(P<0.05);Elastase在腹横筋膜的表达为A组>B组>C组(P<0.05)。结论腹股沟疝患者弹性蛋白合成减少,弹性蛋白酶降解增强影响了腹横筋膜的强度,并进一步影响腹股沟疝的发生和发展,尤其在直疝中更为明显。

【Abstract】 Objective To investigate the expression of Tropoelastin(TE), Lysyl oxidase-like 1( LOXL-1) and Elastase in the transversalis fascia of patients with inguinal hernia. Methods Sixty patients with inguinal hernia were divided into two groups with 30 patients in each group: Direct hernias group(group A) and Indirect hernias(group B). Another 30 patients without hernia disease were selected as control group(group C). All samples were obtained from transversalis fascia in the 3 groups.The protein expression levels of TE,LOXL-1 and Elastase in transversalis fascia were detected by immunohistochemical staining and Western blot analysis. Results There were no significant differences of general conditions among the 3 groups(P>0.05).Both immunohistochemical detection and Western blot analysis showed that the expression levels of TE and LOXL-1 in transversalis fascia were as follows : Group C>B>A(P<0.05). The expression levels of Elastase in transversalis fascia were as follows : Group A>B>C(P<0.05). Conclusion Decreased in synthesis of elastin and enhanced elastase degradation may reduced the strength of the transverse fascia, and contribute to the development of inguinal hernia, especially in direct hernia.
