白藜芦醇在大鼠失血-内毒素诱导的多器官功能障碍中的肺保护作用

【摘要】 目的探讨白藜芦醇(Res)在失血-内毒素二次打击诱导的大鼠多器官功能障碍(MODS)模型中的肺保护作用。方法大鼠30 min内放血2 ml/100 g体重,4 h后腹腔内注射内毒素(5 mg/kg),随机将大鼠分为对照组、盐水组及白藜芦醇组,分别予以生理盐水或20 mg白藜芦醇预处理。采用血气分析仪检测动脉血气;HE染色观察肺部病理变化;伊文思蓝(EB)法检测肺微血管通透性;湿干重比(W/D)检测肺水肿;TUNEL染色检测凋亡细胞数量;蛋白免疫印迹法检测BAX及Bcl-2表达;荧光法检测Caspase-3活性。结果与对照组比较,盐水组中动脉血气明显降低,HE染色见肺泡壁增厚、大量中性粒细胞浸润,肺组织中EB含量及W/D明显增加,差异均有统计学意义(P<0.05),而上述改变在白藜芦醇组中均得到改善,差异有统计学意义(P<0.05);与对照组比较,盐水组中细胞凋亡、Caspase-3活性、BAX表达显著提高,Bcl-2表达降低,差异均有统计学意义(P<0.05),Res治疗可以明显抑制肺组织改变,差异有统计学意义(P<0.05)。结论白藜芦醇可以通过抑制细胞凋亡而减轻失血-内毒素诱导的急性肺损伤。

【Abstract】 Objective To investigate the protective effect of Resveratrol(Res)..against lung injury in hemorrhagelipopolysaccharide induced multiple organ dysfunction syndrome(MODS) in rats. Methods Rats were bled 2 ml / 100 g weight,and 4 hours later intraperitoneally injected lipopolysaccharide 5 mg / kg weight to induce MODS.Rats were randomly divided into the control group,normal saline(NS) group and Res group(20 mg / kg weight). Then rats were correspondingly treated by NS or Res after the double-strike.Pathological changes was detected by hematoxylin-eosin(HE) staining,arterial PO2by blood gas analysis, lung vascular permeability by Evans blue(EB) method, interstitial edema by wet-to-dry weight ratio(W / D), the number of apoptotic cells by TUNEL staining, Bax,Bcl-2 expression by Western-blot,Caspase-3 activity by Caspase-3 fluorometric assay kit. Results Rats in NS group exhibited significant acute lung injury(ALI) characteristics:deterioration of histopathological features, increased in W / D ratio and EB concentration, and decreased in PO2(P<0.05, compared with control group). These characteristics could be attenuated by Res(P<0.05).Compared with control group, number of TUNEL-positive cells were increased, caspase-3 was activated, Bax was upregulated and Bcl-2 was down-regulated in NS group(P <0.05);these could also be prevented by Res treatment(P <0.05). Conclusion Res treatment ameliorates hemorrhage-lipopolysaccharide induced ALI via apoptosis depression.
