硒拮抗铅上调SH-SY5Y和PC12细胞β-淀粉样蛋白表达的研究
Effects of selenium on lead-induced alterations in Aβ production in SH-SY5Y and PC12 cells
【摘要】 目的研究硒能否拮抗铅上调SH-SY5Y和PC12中β-淀粉样蛋白40和β-淀粉样蛋白前体蛋白表达。方法用醋酸铅和硒蛋氨酸联合染毒SH-SY5Y和PC12细胞,CCK8试剂盒测定细胞存活率,蛋白印迹法和酶联免疫法测定β-淀粉样蛋白前体蛋白和β-淀粉样蛋白40表达。结果单独染铅48 h后,SH-SY5Y和PC12两种细胞中β-淀粉样蛋白40和β-淀粉样蛋白前体蛋白表达均增加,且随着铅剂量的增加表达量增加;与高剂量染铅组相比,铅硒联合作用组β-淀粉样蛋白40和β-淀粉样蛋白前体蛋白表达下降,且随着硒剂量的增加表达量减少。结论硒能拮抗铅上调β-淀粉样蛋白表达的作用。
【Abstract】 Objective To study the antagonistic effect of selenium on lead-induced alterations in beta-amyloid(Aβ) and beta-amyloid precursor protein(APP) production in SH-SY5Y and PC12 cells. Method After co-incubation with lead acetate and selenium methionine(SeMet), the SH-SY5Y and PC12 cells survival was tested by CCK8 kit, the expression of APP and Aβ was detected by Western-blot and ELISA. Results Lead incubation alone induced an increase in the expression of Aβ and APP compared to that of control group in SH-SY5Y and PC12 cell after treated for 48 h in dose dependent manor. SeMet could inhibit up-regulation of beta-amyloid(Aβ) and beta-amyloid precursor protein(APP) production induced by lead in SH-SY5Y and PC12 cells in dose dependent manor. Conclusion Selenium has the antagonistic effect on lead-induced over-accumulation of Aβ.
