温度骤变对栉孔扇贝全血细胞和颗粒血细胞数量的影响
Influence of Temperature Shock on the Numerical Variation of Scallop Hemocytes and Granulocytes
【摘要】 血细胞及其亚群的数量是栉孔扇贝(Chlamys farreri)的重要免疫指标之一。本文研究了栉孔扇贝在17℃下,分别经水温突变(11、23和28℃)和不同温度(5、11、17和23℃)干露处理后,应用以抗栉孔扇贝全血细胞单抗和颗粒血细胞单抗分别为第一抗体的间接酶联免疫吸附实验和流式细胞术测定全血细胞数量、颗粒血细胞数量及比例的变化。结果表明:水温骤变全血细胞和颗粒血细胞数量变化显著,全血细胞数量11、23℃组均先升高后逐渐趋于对照组,28℃组一直显著低于对照组;颗粒血细胞数量及其比例11℃组显著增加并始终高于对照组,23℃组显著降低后逐渐恢复至对照水平,28℃组始终低于对照水平。不同温度干露后,全血细胞和颗粒血细胞数量均显著降低,海水恢复后,5℃组较11、17℃组更接近对照组,23℃组干露12h扇贝全部死亡。研究表明:全血细胞及颗粒血细胞数量对温度变化的反应敏感;较低水温有利于细胞数量的提升,较高水温下细胞数量下降显著;低温干露后细胞数量恢复较好,高温干露扇贝存活率差。
【Abstract】 In this study,scallop(Chlamys farreri)was shocked with different temperatures by transferring them from 17℃to 11,23and 28℃,respectively,or air exposing them to 5,11,17and 23℃,respectively.The hemocytes and granulocytes were counted with enzyme-linked immunoadsorbent assay(ELISA)by using monoclonal antibody against hemocytes or granulocytes as the primary antibody separately.The percentage of hemocytes and granulocytes was determined with flow cytometry.The results showed that total hemocyte count(THC)increased significantly at 11and 23℃,but declined apparently at 28℃;while granulocyte count(GC)was significantly higher at 11℃than at 23and 28℃ with the minimum detected at 28℃.The percentage of granulocytes(PG)varied in the same way as GC did.GC and PG were always lower than those of control group.THC,GC and PG descended significantly after air exposure at 5,11and 23℃.It was found that THC,GC and PG of scallop air exposed at 5℃ were the highest,which were recovered when scallop were transferred back into seawater.No scallop survived air exposure at 23℃for 6h.In conclusion,the number of hemocytes and granulocytes was sensitive to temperature shock,being lower at 28℃than at other temperatures;air exposure significantly influenced hemocyte and granulocyte count,and the survival rate at 23℃ was the lowest.
