大蒜素对人卵巢癌SKOV3/DDP细胞增殖与凋亡的影响及机制
Diallyl trisulfide inhibits proliferation and triggers apoptosis in human ovarian cancer SKOV-3/DDP cell
【摘要】 探讨大蒜素(DT)对人上皮性卵巢癌SKOV-3/DDP细胞的增殖抑制及诱导其细胞凋亡的分子机制。用人上皮性卵巢癌耐顺铂细胞株SKOV-3/DDP细胞随机分为4组:①正常对照组;②DT组:于培养基中加入DT(终浓度分别为5、10、20、40μg·mL-1)分别作用细胞24、48和72h;③顺铂(DDP)组(5μg·mL-1);④DT+DDP组(20μg·mL-1 DT和5μg·mL-1 DDP)。MTT比色法检测细胞存活率,流式细胞术检测细胞凋亡率,RT-PCR和Western blot分别检测基因bax、bcl-2的mRNA和蛋白表达。与正常对照组比较,DT组SKOV-3/DDP细胞增殖抑制率与凋亡率均有显著的增加(P<0.05),且作用呈剂量和时间依赖性,Bax mRNA和蛋白表达上调,Bcl-2mRNA和蛋白表达下调(P<0.05);与DT或DDP组比较,DDP+DT组的细胞生长抑制率及凋亡率均有显著增加(P<0.05),且Bax mRNA及蛋白表达上调,Bcl-2mRNA和蛋白表达下调的程度大于单用组(P<0.05)。大蒜素对SKOV-3/DDP细胞有显著的抑制增殖和促凋亡的作用,增加SKOV-3/DDP细胞对顺铂的敏感性,其机制可能与大蒜素调控Bax/Bcl-2蛋白表达有关。
【Abstract】 Aim:To investigate the effects of Diallyl Trisulfide(DT)on proliferation and apoptosis in human epithelial ovarian cancer SKOV-3/DDP cells and its mechanism.Cultured cisplatin-resistant human epithelial ovarian cancer SKOV-3/DDP cells were randomly divided into four groups:the control,DT-treated groups(5,10,20,40μg·mL-1),DPP-treated group(5μg·mL-1),and DPP(5μg·mL-1)+DT(20μg ·mL-1)group,which were incubated for 24,48and 72hrespectively.Detected the cell viability by(MTT) and the apoptosis rate of cancer cells by flow cytometric analysis.Detected the mRNA and protein expressions of bax and bcl-2in the ovarian cancer SKOV-3/DDP cells by RT-PCR or by Western blot respectively.Compared with the control,DT could inhibit the proliferation and induce the apoptosis in SKOV-3/DDP cells in a dose and time-dependent manner(p< 0.01),up-regulate the mRNA and protein expressions of gene bax,and down-regulate the mRNA and protein expressions of gene bcl-2.Compared with the DT or DDP,inhibit rate and apoptosis rate of the cancer SKOV-3/DDP cells in the DDP+DT(P<0.05)were increased.DT is able to inhibit proliferation of the human epithelial ovarian cancer SKOV-3/DDP cells and enhance the cell chemosensitization to DDP.Bax/Bcl-2proteins might be involved in this process.
