MTA1蛋白激活与宫颈癌上皮间质转化的相关性研究

【摘要】 目的探讨MTA1与宫颈癌上皮间质转化的相关性。方法实验采用免疫组化方法检测了101例宫颈鳞状细胞癌中MTA1,E-cadherin和Vimentin的表达,分析MTA1蛋白激活与E-cadherin和Vimentin表达的关系。结果 MTA1,Vimentin及E-eadherin在宫颈癌组织中阳性表达率分别为69/101(68.3%),70/101(69.3%)和101/101(100.0%)。MTA1表达与Vimentin表达呈正相关,与E-cadherin表达呈负相关(均P<0.05)。Vimentin表达与宫颈癌组织分级、淋巴结转移有关(均P<0.05),而E-cadherin的表达与淋巴结转移有关(P<0.05)。结论①MTA1在宫颈癌中的表达与组织分化程度、临床分期及淋巴结转移密切相关。②E-cadherin的表达与淋巴结转移有关,MTA1表达水平与年龄、肿瘤大小无明显相关性。③MTA1能调控宫颈癌细胞生物学行为及EMT过程,加强宫颈癌细胞恶性生物学行为,导致侵袭和转移能力的增强。

【Abstract】 Objective To investigate the expression and correlation among signal transducer and activator of MTA1 and E-cadherin and Vimentin in cervical cancer,and analyze their relationship with epithelial-mesenchymal transition and tumor infiltration and metastasis.Methods In this study,immunohistochemical method was applied to detect the expression of MTA1 in 101 cervical squamous carcinoma and cervical lymph node metastasis cancer tissues,which was aimed to investigate the impact and mechanism of MTA1 on invasion and metastasis in cervical cancer cells and epithelial-mesenchymal transition(EMT).Results The expression rate of MTA1 was 68.3%in cervical lymph node metastasis cancer tissues,Vimentinin was 68.3% and E-cadherin was 100.0%.The expression of MTA1 was significant correlation with E-cadherin and Vimentinin(P <0.05).MTA1 and Vimentinin was prominently expressed within the higher histology grades,metastasis of lymph nodes(P < 0.05).The expression of E-cadherin was significantly associated With metastasis of lymph nodes(P < 0.05).The expression of pSTAT3,E-cadherin and Vimentinin were not significantly associated with age,tumour size.Conclusion The expression rate of MTA1 is significant related to tissue differenation degree,clicinal stage and lymph metastasis.In addition to,the expression of E-cadherin was closedly to lymph node metastasis while the expression level of MTA1 had no significant correlation with age and tumor size.Moreover,MTA1 can regulate biology behavior of cervical cancer and EMT process,leading to the improvement of the ability of invasion and metastasis.
