GC-MS结合内标法分析冬虫夏草中的脂肪酸含量
Quantitative analysis of fatty acids in Cordyceps sinensis by GC-MS with internal standard method
【摘要】 目的建立一种气相色谱—质谱联用(GC-MS)同时测定冬虫夏草中9种脂肪酸的检测方法。方法冬虫夏草中的脂肪酸用10%硫酸—甲醇衍生化,产物由二氯甲烷萃取后用GC-MS检测。结果通过优化前处理条件,9种脂肪酸的含量与峰面积线性关系良好(0.9992~0.9999),方法的回收率高(83.75%~0102.05%),重复性好(RSD<3.0%)。对青海四个地区及西藏某地区的冬虫夏草进行分析,共检测出8种脂肪酸。结论该方法准确、操作简单,可用于定量分析冬虫夏草中的脂肪酸含量。
【Abstract】 Objective To develope a method for the quantitative analysis of fatty acids in cordyceps sinensis by gas chromatography-mass spectrometry.Methods The fatty acids contained in Cordyceps sinensis were methyl-esterified in 10%H2SO4-CH3OH,followed by extraction with methylene dichloride,and determined by gas chromatography-mass spectrometry.Results The method has high recovery(83.75%-102.05%) and good reproducibility(relative standard deviation < 3.0%).For its superiority,cordyceps sinensis from four regions in Qinghai and one region in Tibet were analyzed,and 8 kinds of fatty acids were separated and identified.Conclusion This method is accurate,simple fast and can be used in the quantitative analysis of fatty acids in Cordyceps sinensis.
