化疗期间不同时间段肿瘤患者CIK细胞抗肿瘤作用的体外研究

【摘要】 目的:研究同一肿瘤患者在化疗期间不同时间段自体CIK细胞对不同癌细胞株杀伤活性的差异。方法:分离化疗期间不同时间段肿瘤患者外周血单个核细胞(PBMC),加入细胞因子,体外诱导CIK细胞,用MTT法检测其对不同时间段不同癌细胞株的杀伤活性及随化疗次数变化的趋势,杀伤活性比较采用方差分析。结果:化疗期间不同时间段CIK细胞对肺癌(A549)、肠癌(HCT-116)、鼻咽癌细胞珠(CNE2)的杀伤活性比较:同一肿瘤患者的CIK细胞随着化疗次数的递增,对肺癌细胞株、肠癌细胞株的杀伤活性逐渐降低,不同时段差异具有统计学意义(P<0.001),鼻咽癌细胞株杀伤活性无规律性变化(P>0.05);同一化疗时间段,CIK细胞对不同癌细胞株杀伤活性的比较:对肺癌细胞株的杀伤活性最强,对鼻咽癌细胞株的杀伤活性最差。结论:化疗联合自体CIK细胞在化疗不同时间段对不同癌细胞株的杀伤活性有差异。

【Abstract】 Objective: To investigate the effect of CIK cells at different chemotherapy phase on a variety of tumor cell lines. Methods: PBMC was harvested from patients at diverse phases of chemotherapy, and then differentiated into CIK cells with induction of cytokine. The activity of CIK cells on tumor cell lines was determined by MTT assay, and data was assayed by ANOVA. Results: The activity of CIK cells from patients at diverse phases of chemotherapy on A549 and HCT-116 decreased significantly, while this difference was not observed on CNE2. Meanwhile, at the same phase of chemotherapy, A549 was the most sensitive cell to CIK cell treatment, while CNE2 was the least. Conclusion: The activity of chemotherapy combined with CIK cells therapy at different phases varies on tumor cell lines.
