骨髓间充质干细胞移植联合电针治疗对脑出血大鼠脑组织中BMP2表达的影响
Effect of combined treatment of mesenchymal stem cells transplantation and electric acupuncture on the expression of BMP2 in rats with intracerebral hemorrhage
【摘要】 目的:探讨骨髓间充质干细胞(MSCs)移植和电针(Ea)联合治疗大鼠脑出血性损伤对脑组织中骨形态发生蛋白-2(BMP2)表达的影响。方法:100只SD大鼠尾壳核处注射肝素和胶原酶建立脑出血动物模型,并随机分成生理盐水组(Ns组)、电针组(Ea)、MSCs组、MSCs移植联合电针组(MSCs+Ea)共4组,每组25只。术后第3 d分别给予注射生理盐水、电针刺激、MSCs移植、MSCs移植联合电针刺激,每组又根据治疗时间的不同分为1、3、5、7、14 d五个亚组,每个亚组5只。到相应时间点时麻醉处死大鼠,取脑制作冰冻切片,采用免疫组化方法检测BMP2的表达。结果:对各组BMP2阳性细胞数比较显示:MSCs+Ea组各时间点比Ns组相应时间点增多(P<0.05),在1、5、7 d时比Ea组上升(P<0.05),在1 d和14 d时比MSCs组增加(P<0.05);MSCs组比Ns组在3 d和5 d时增多,比Ea组在1 d和5 d时上调(P<0.05)。Ns组和Ea组相比,P>0.05,差异无统计学意义。结论:MSCs移植和电针联合治疗比单纯的MSCs移植或电针刺激更能促进脑出血大鼠脑组织中BMP2的表达。
【Abstract】 Objective: To explore the effect of combined treatment of mesenchymal stem cells(MSCs)transplantation and electric acupuncture(Ea) on the expression of BMP2 in rats with intracerebral hemorrhage(ICH). Methods: 100 rat ICH models were set up by injecting collagenase and heparin into the caudate putamen nucleus in rats. The rats were randomly divided into four groups(n = 25): Ns group, Ea group, MSCs group and MSCs + Ea group. Each group was divided into 1 d,3 d,5 d,7 d and 14 d subgroups. The rat brains were took out and frozen sections were processed. The expression of BMP2 was detected by immunohistochemistry. Results:BMP2 expression of MSCs + Ea group was higher than that of Ns group at all subgroups(P < 0.05), higher than that of Ea group at 1 d, 5 d and 7 d(P < 0.05) and higher than MSCs group at 1d and 14 d(P < 0.05). The BMP2of MSCs group raised remarkably compared with Ns group at 3 d and 5 d(P < 0.05), and more remarkable than that of Ea group at 1 d and 5 d(P < 0.05). BMP2 cells had no significant difference between Ns group and Ea group(P > 0.05). Conclusion: The combined treatment of MSCs transplantion and electric acupuncture promotes the expression of BMP2 in ICH rats.
