栽培蓝莓-园蓝的抗氧化活性及其活性化合物的定性定量

【摘要】 为测定栽培蓝莓-园蓝果实中的抗氧化活性及其总花色苷含量,以贵州产园蓝鲜果为原料,经甲醇处理、大孔吸附树脂分离得出抗氧化活性最好的部位。然后对其进行了制备分离得到一单体化合物,经1 H-NMR、13 C-NMR、ESI-MS进行了结构鉴定,利用Rp-HPLC进行了该化合物的含量测定。以该化合物为对照品,对园蓝果实中总花色苷含量进行了定量分析。研究表明:贵州产园蓝提取物富含花青素部分1mg/mL浓度的抗氧化活性与等浓度的维生素C的活性相当,鉴定的单体化合物为锦葵色素-3-O-葡萄糖,其纯度达96%,园蓝鲜果中总花青素平均含量为627.7mg/100g。指出了园蓝鲜果花色苷含量高,主成分为锦葵色素-3-O-葡萄糖,花青素具有较好的抗氧化活性。建议每人每天食用2g园蓝鲜果作为人体补充抗氧化剂的来源。

【Abstract】 The Objective of the article is to determine the antioxidant activity and quantification of anthocyanin in the cultivated blueberry--gardenblue.The Method of the experiment is that taking the fresh fruit of gardenblue cultivated in Guizhou province as raw material and extracting the part of the best antioxidant activity through methanol treatment and macroreticular resin.Then,the preparative separations of part of the best antioxidant activity gets monomeric compound which is tested 1 H-NMR,13 C-NMR and ESI-MS,and quantitative analysis of the monomeric compound is done based on Rp-HPLC.Considering the compound as a comparison product,the experiment makes a quantitative analysis of total anthocyanidins.The results show that in the extracted part which contains anthocyanidins,1mg/mL concentration’s antioxidant activity is equivalent to that of the same concentration of Vitamin C.The appraised monomeric compound is malvidin-3-O-glucose whose purity reaches 96%.The average content of total anthocyanin of gardenblue’s fresh fruits is 627.7mg/100 g.The conclusion is that the gardenblue contains the richest anthocyanin whose main component is malvidin 3-O-glucose,and,anthocyanin shows a better antioxidant activity.Therefore,the article advises that people should consume 2g fresh fruits of gardenblue everyday as the source of antioxidant.
