抗肝衰竭良方——三黄茵赤汤的急毒试验

【摘要】 目的:研究抗肝衰竭良方—三黄茵赤汤的急性毒性,为临床安全使用该方提供实验依据。方法:用最大给药量——150g/(kg/d)的三黄茵赤汤给小鼠灌胃7 d,观察小鼠的毒性反应,记录小鼠的死亡情况。检测小鼠血清中谷草转氨酶(AST)和谷丙转氨酶(ALT)的含量,病理切片观察肝脏的损伤程度。结果:实验小鼠未出现毒性反应,与对照组比较,体重变化差异不显著(P>0.05),与对照组比较,AST和ALT的含量无统计学差异(P>0.05),无肝脏病理损伤情况,最大给药量相当于60 kg体重成人临床推荐剂量的82倍。结论:三黄茵赤汤最大给药量对实验小鼠肝脏无毒性,三黄茵赤汤临床剂量服用安全。

【Abstract】 Objective: To study the acute toxicity of Sanhuang Yinchi Decoction( SHYC) with resistance to liver failure in order to provide the experimental basis for clinical use. Methods: SHYC decoction at the concentration of 150 g /( kg /d) was orally given to mice for 7 days. We observed toxic reaction and recorded the number of dead mice. The concentrations of both AST and ALT in mice were determined. Liver slices were used to observe damage of liver. Results: The obvious toxic reaction was not observed in experimental group. There was no significant difference in body weight to compare with the control group( P > 0. 05).There was no significant difference in AST and ALT to compare with the control group( P > 0. 05) and no pathological injury of liver. The maximum administration dose was 82 times the clinical dose. Conclusion: The maximum administration dose of SHYC decoction can not induce liver damage in mice and it is safe in clinic.
