银屑病患者皮损中转化生长因子-β1、-β1受体及其信号转导蛋白Smad2、3的表达
The expression of TGF-β1、TGF-β1 receptors and Smad 2、3 in psoriatic lesions
【摘要】 目的:观察银屑病患者皮损与正常人皮肤中T细胞转化生长因子(TGF)-β1、TGF-β1受体(TGF-β1R I、TGF—β1RⅡ)及其信号转导蛋白Smad2、3的表达,分析TGF—β1信号转导通路在银屑病发病机制中的作用。方法:采用Real-time PCR(RTPCR)法检测20例银屑病患者皮损及10例正常人皮肤中TGF—β1、TGF—β1受体(I、Ⅱ)、Smad(2、3)mRNA的表达,用western blot杂交法检测TGF—β1、Smad2、3的表达。结果:银屑病患者皮损中TGF—β1、TGF—β1R I、TGF—β1RⅡ及其信号转导蛋白smad2、3的mRNA表达明显低于正常对照组(P<0.01),TGF—β1和Smad2、3的蛋白表达明显低于正常对照组(P<0.01)。结论:T细胞TGF-β1信号转导通路的低表达可能有助于银屑病患者表皮过度增殖状态的发生和发展。
【Abstract】 Objective:To investigate the expression of TGF-β1,TGF-β1 receptors(TGF-β1R Ⅰ and TGF-β1R Ⅱ) and signaling proteins,Smad2 and Smad3,in psoriatic lesions and normal human skin,and to analyze their roles in the pathogenesis of psoriasis.Methods:Expression levels of TGF-β1,TGF-β1 receptors,Smad2 and Smad3 mRNA were assessed by Real-time PCR(RT-PCR) in 20 psoriatic patients and 10 normal controls.Western blot was used to assess the expression levels of TGF-β1,Smad2 and Smad3.Results:mRNA levels of TGF-β1,TGF-β1 receptors(TGF-β1R Ⅰ and TGF-β1RⅡ),Smad2 and Smad3 were significantly down-regulated in psoriatic lesions compared with normal controls(P<0.01).The protein levels of TGF-β1,Smad2 and Smad3 were also significantly down-regulated in psoriatic lesions compared with normal controls(P<0.01).Conclusion:The down-regulated TGF-P,signaling pathway in psoriatic lesions might contribute to the formation of hyperplasic epidermis in psoriasis.
