瘦素和瘦素受体mRNA表达与哮喘和肥胖的关系
Associations of leptin and leptin receptor mRNA expression to asthma and obesity
【摘要】 目的探讨瘦素(LEP)和瘦素受体(LEPR)mRNA表达与哮喘和肥胖的关系。方法采用实时荧光定量PCR(RT-qPCR)方法测定各组PBMC LEP和LEPR mRNA表达水平,分析LEP mRNA与血浆瘦素浓度、EOS%、IgE和FEV1占预计值(%)的关系。结果哮喘、肥胖、哮喘合并肥胖组的PBMC LEP mRNA表达量与健康对照组相比明显升高(P均<0.01),中、重度哮喘与轻度哮喘相比明显升高(P均<0.05);肥胖组LEPR mRNA表达量与其余各组相比明显升高(P均<0.01);哮喘组PBMC LEP mRNA表达量与血浆LEP浓度呈正相关(r=0.211,P=0.012),与FEV1占预计值(%)成负相关(r=-0.314,P=0.043),与EOS%、IgE无相关关系(r=0.002、0.001,P=0.58、0.96)。结论 LEP mRNA表达升高可能参与哮喘和肥胖的发生,并且导致较为严重的哮喘表型,其机制可能与血浆瘦素浓度增高有关,其相关的哮喘是非嗜酸细胞性、非过敏性的;LEPR mRNA高表达参与肥胖发生,但与哮喘无关。
【Abstract】 Objective To investigate the associations of leptin and leptin receptor mRNA expression in peripheral blood mononuclear cells to asthma and obesity. Methods Real-time fluorescence quantitative polymerase chain reaction( RT-qPCR) was used to detect the expression of relative levels of LEP and LEPR mRNA in PBMC.The relationships of LEP and LEP mRNA to IgE,EOS% and FEV1% were analyzed. Results There were statistical differences in the levels of leptin mRNA among the asthma,obesity,asthma-combined obesity and the health control groups( all P < 0. 01). The mod-severe asthma patients had a higher leptin mRNA expression levels than mild asthma patients did( all P < 0. 01). There were statistical differences in the levels of leptin receptor mRNA among the obesity and asthma,asthma-combined obesity,and health control groups. The leptin mRNA level was positively correlated with plasma leptin concentration( r = 0. 211,P = 0. 012),and negatively correlated with FEV1%( r =- 0. 314,P = 0. 043). There was no correction with plasma total IgE and eosinophile counts( r = 0. 002,0. 001,P= 0. 58,0. 96). Conclusion The high expression of PBMC leptin mRNA may induce asthma and obesity and the type of asthma induced is more severe and non-eosinophilic and non-allergic. The possible mechanism is higher mRNA expression induce PBMC to produce more leptin then contribute to the inflammatory reaction of asthma. The high expression of PBMC leptin receptor mRNA induces obesity while has no influence on asthma.
