PDZK1在肾癌中的表达及意义

【摘要】 目的:研究PDZK1在不同分化程度肾癌中表达,探讨其与肾癌分化程度的相关性。方法:通过免疫组织化学(ABC法)对60例不同分化程度肾癌和20例肾脏良性病变的病理组织检测PDZK1的表达情况,并利用RT-PCR技术对20例新鲜肾癌组织和10例肾脏良性病变组织检测PDZK1的mRNA表达。结果:PDZK1阳性率在肾癌组表达为85.0%,在肾脏良性病变组表达为30%(P<0.05),在肾癌组分级G1、G2、G3阳性表达率分别为61.5%、63.2%和86.7%,组间两两比较(P>0.05)。PDZK1的mRNA在肾癌组表达为0.95±0.055,在肾脏良性病变组表达为0.57±0.043(P<0.05);PDZK1的mRNA在肾癌组分级G1、G2、G3表达分别为0.72±0.044、0.831±0.090和1.00±0.050,组间两两比较(P<0.05)。结论:PDZK1在肾癌组织中高表达,随恶性度增高表达呈上升趋势,在肿瘤恶性进展中具有重要的作用。

【Abstract】 Objective: To research the expression of PDZK1 domain containing protein 1(PDZK1) in rena carcinoma tissue,and toinvestigate the correlation of PDZK1 with renal carcinomain differentgrades. Method: Expression of proteins of PDZK1 in sixty cases of different grade renal carcinoma pathological tissue and 20 cases of benign kidney disease pathological tissue were detected by immunohistochemistry(ABC), and mRNA expression of PDZK1 in 20 cases of surgical fresh tissue with renal carcinoma and 10 cases of with benign kidney disease fresh tissue were also detected by reverse transcription polymerase chain reaction(RT- PCR). Results: The positive expression rate of PDZK1 is 85.0% in different grade renal carcinoma pathological tissue, and 30% in benign kidney disease tissues(P < 0.05), 61. 5%, 63. 2%, 86. 7% in G1, G2, G3grade(P> 0.05). The expression of mRNAof PDZK1 is 0.95± 0.055 in renal carcinoma group, 0.57± 0.043 in benign kidney disease group(P <0.05). The expression of mRNAof PDZK1 is 0.72± 0.044,0.831± 0.090, 1.00± 0.050 in G1,G2,G3grade of renal carcinoma respectively(P <0.05). Conclusion: High expression of PDZK1 in renal carcinoma was followed by the degree of tumor progress,which indicated that the PDZK1 plays an important role in the progress of renal carcinoma malignant.
