【摘要】 目的研究Th17细胞相关的主要细胞因子IL-17、IL-6和TGF-β在丙型肝炎患者外周血中的表达情况并分析与患者临床特征的相关性,初步探讨Th17细胞在丙型肝炎病理过程中的作用.方法收集54例丙型肝炎患者做为阳性组,并以40例正常志愿者为对照组.采用ELISA方法检测外周血血清中IL-17、IL-6、TGF-β的含量,同时测定HCV-RNA和肝功能指标ALT、AST、TBil、ALB等.结果研究发现,丙型肝炎患者外周血中IL-17、IL-6和TGF-β水平明显高出对照组(P=0.004,P=0.018,P=0.025).IL-17的升高与IL-6和TGF-β相关(r=0.664,P=0.00,r=0.511,P=0.00).IL-17水平与ALT,AST和HCV-RNA相关(r=-0.52,P=0.01;r=-0.282,P=0.005,r=-0.209,P=0.037),但是TGF-β、IL-6均与ALT,AST或HCV-RNA没有明显的相关性,IL-6浓度与TGF-β有关(r=0.807,P=0.00).结论丙型肝炎患者外周血中Th17相关细胞因子表达增高,特别是IL-17与患者ALT、AST以及病毒载量相关,而且增高的IL-17与血清IL-6、TGF-β水平密切相关.提示Th17细胞参与了丙型肝炎的免疫学发病机制.更多还原

【Abstract】 Objective To investigate the expression of serum IL-17, IL-6 and TGF-β in chronic hepatitis C virus infection patients and explore the correlation between IL-17, IL-6, TGF- β and clinical characteristic of hepatitis C patients.Methods 54 cases of patients with chronic hepatitis C and 40 healthy volunteers were enrolled. The expression of IL-17,IL-6 and TGF- β in serum were evaluated by ELISA;ALT,AST,ALB and TBil were determined by ABBOTT AEROSET Ci16200 system, HCV-RNA were measured by RT-PCR respectively. Results Compared with healthy controls,the levels of IL-17, IL-6 and TGF-βwere all increased significantly in hepatitis C patients serum(P=0.004,P=0.018,P=0.025).Correlation analysis showed the elevated IL-17 was related to IL-6(r=0.664,P=0.00) and TGF-β(r=0.511,P=0.00). IL-6 was also related to TGF-β(r=0.807,P=0.00). Increased IL-17 was associated with ALT,AST and HCV-RNA(r=-0.52,P=0.01; r=-0.282,P=0.005,r=-0.209,P=0.037), But no obvious correlations between TGF-βand ALT,AST or HCV-RNA(r=-0.104,P=0.424,r=-0.034,P=0.871,r=-0.167,P=0.74)were observed,It is similar to IL-6 and ALT, AST or HCV-RNA(r=-0.13,P=0.951,r=-0.118,P=0.576, r =-0.083, P =0.694). Conclusion All the results suggest that Th17 cells participate in the immunological pathogenesis of chronic hepatitis C.更多还原
