【摘要】 目的探讨颗粒蛋白前体(progranulin,PGRN)在胃溃疡愈合中的表达变化及其作用。方法采用免疫组织化学染色和RT-PCR方法对32例胃溃疡患者愈合前后胃黏膜组织中PGRN的表达进行检测。结果免疫组化结果显示,PGRN主要表达于胃黏膜上皮及腺上皮细胞胞质中,溃疡愈合后胃黏膜组织中PGRN的表达水平与愈合前相比明显减弱(t=9.026,P<0.01)。RT-PCR检测结果显示PGRN mRNA的表达与其蛋白质的改变相一致(P<0.01)。结论 PGRN可能作为一种重要的促进因子,通过促进胃黏膜上皮增生,促进胃溃疡愈合。更多还原

【Abstract】 Objective To explore the role of progranulin( PGRN) in healing of gastric ulcers via investigating the expression and distribution of PGRN in gastric mucosa. Methods Expression of PGRN in gastric mucosa in 32 patients with gastric ulcer was examined by SP immunohistochemistry stain and reverse transcription polymerase chain reaction( RT-PCR) before and after the treatment. Results PGRN was mainly found in gland cells of gastric mucosa,and compared with before treatment,its expression level in stained tissue after treatment was significantly decreased( t = 9. 026,P < 0. 01). The mRNA expression level of PGRN before and after treatment changed in a similar way as that of PGRN protein.
