【摘要】 目的探讨随机扩增多态性DNA(RAPD)分析法在在幽门螺杆菌(HP)菌株鉴定中的应用价值。方法选取该院65例HP感染的慢性胃炎或消化性溃疡患者,获取胃窦、胃体部黏膜进行HP培养,提取及纯化HP DNA后进行PCR反应,并采用RAPD分析HP基因指纹图谱;以同一HP菌株验证RAPD可重复性;65例患者均进行6个月以上的HP治疗,治疗后4~6周评估HP治愈情况;对复发病例再次进行RAPD分析,观察RAPD指纹图谱变化。结果 65株菌株PAPD指纹图谱各不相同,但同一菌株传4代后,原代株与传代株进行PCR扩增,RADP指纹图谱完全一致。6例HP治疗后复发,3例RAPD指纹图谱与治疗前基本一致;3例产生不同RAPD指纹图谱。结论 RAPD分析法适用于HP基因多态性鉴定,是临床研究HP感染的实用方法,可用于HP复发和再感染鉴定。更多还原

【Abstract】 Objective To investigate application value of random amplified polymorphic DNA analysis in the identification of HP.Methods 65patients with peptic ulcer or chronic gastritis,which confirm as HP infection,get orget gastric body mucosa carried HP culture,after HP DNA extraction and purification,and analysis using HP RAPD genetic fingerprinting;strains of the same HP RAPD verify repeatability,65patients underwent six months or more HP eradication therapy,assess the situation Hp eradication 4-6weeks after treatment;recurrence again on RAPD analysis,the changes of RAPD fingerprint were observed.Results 65strains PAPD fingerprints are different,but RADP fingerprints of the original generation plants and extend the plant after 4passages is same.Three cases is consistant in RAPD fingerprinting befor and after treatment,3cases have different RAPD fingerprints in six cases of relapse after HP eradication.Conclusion RAPD analysis of genetic polymorphisms identified for HP,is a useful tool for clinical studies,it can be used to identify HP relapse and reinfection.更多还原
