行为学训练对大鼠内源性神经干细胞增殖及Wnt3a蛋白表达的影响

【摘要】 目的:检测Wnt3a在血管性痴呆海马组织神经干细胞中的表达及穿梭箱训练的干预作用,探讨影响神经干细胞增殖分化的分子调控机制。方法:采用大脑中动脉栓塞(MCAO)制作大鼠血管性痴呆模型,用免疫组织化学检测海马神经干细胞BrdU、Wnt3a的表达及穿梭箱训练的干预作用。结果:随着脑缺血再灌注第3天缺血海马神经元BrdU阳性细胞明显增多,第7天达高峰,且高于假手术组大鼠。Wnt3a反应产物在脑缺血再灌注第3天较正常组增多,持续高水平表达到第7~14天,随缺血再灌注时间的进一步延长逐渐减少,高于假手术组大鼠。穿梭箱训练后BrdU、Wnt3a的表达明显增加,明显高于模型组及假手术组。结论:穿梭箱训练促进神经干细胞增殖及缺血脑组织Wnt3a的表达,提示穿梭箱训练促进神经干细胞增殖作用可能由Wnt3a信号介导。

【Abstract】 Objective:To investigate the expression of Wnt3 a expression and hippocampal neurogenesis in a rat model of cerebral ischemia/reperfusion,and to explore the related molecular and behavior training mechanism.Methods:Wistar rats were randomized into training groups,ischemia and reperfusion groups and a control group.Behavior training was performed at day 1 after the ischemia and reperfusion.The model of middle cerebral arteries occlusion(MCAO) was performed with intraluminal filament blockade.The expression of BrdU and Wnt3 a in the hippocampus was detected with immunohistochemical methods.Results:After 3 days,BrdU positive cells in the hippocampus were obviously increased.On day 7,BrdU positive cells were more than those at any time,and higher than the sham rats.Wnt3 a reaction products were increased gradually with the time of cerebral ischemia/reperfusion,reached the peak on 14 day,and then gradually decreased,and higher than sham rats.After training,the expression of hippocampal BrdU and Wnt3 a was apparently increased in each group,higher than the sham and ischemia rats.Conclusion-.Behavior training promotes the expression of Wnt3 a in the hippocampus tissues.These findings indicate that training promotion of neural stem cell proliferation may be mediated by Wnt3 a signaling pathway.
