盐酸埃克替尼对人舌鳞癌Tca8113细胞MMP-9、E-cadherin蛋白表达的影响

【摘要】 目的:研究表皮生长因子受体酪氨酸激酶抑制剂盐酸埃克替尼对人舌鳞状细胞癌Tca8113细胞MMP-9、E-cadherin蛋白表达的影响,探讨其是否可抑制肿瘤的浸润、转移。方法:分别应用0、20μmol/L盐酸埃克替尼处理体外培养的人舌鳞状细胞癌Tca8113细胞,48 h后采用间接免疫荧光联合流式细胞及免疫细胞化学检测盐酸埃克替尼对人舌鳞癌Tca8113细胞MMP-9、E-cadherin蛋白表达的影响。结果:盐酸埃克替尼作用于人舌鳞癌Tca8113细胞后,流式细胞术和免疫细胞化学均显示MMP-9蛋白表达降低,E-cadherin蛋白表达增加。结论:盐酸埃克替尼可降低MMP-9蛋白表达,增加Ecadherin蛋白表达,从而可抑制人舌鳞癌Tca8113细胞的浸润、转移。

【Abstract】 Objective:To study the effect of icotinib on the expression of MMP-9 protein and E-cadherin protein in human tongue carcinoma cell line Tca8113 and explore whether it can inhibit tumor infiltration and metastasis.Methods:Tca8113cells were incubated with icotinib at a dose of 0,20 μmol/L for 48 h.Immunocytochemical method,indirect immunfluorescene and FCM were used to detect the expression of the protein MMP-9,E-cadherin after Tca8113 cells were treated with icotinib.Results:Immunocytochemical,indirect immunfluorescene and FCM results indicated that icotinib decreased the expression of the MMP-9 protein and increased the expression of the E-cadherin protein in Tca8113 cells.Conclusion:Icotinib can decrease the expression of the MMP-9 and increase the expression of the E-cadherin.This result further demonstrates that icotinib can inhibit infiltration and metastasis of human tongue carcinoma cell line Tca 8113.
