CIAPIN1在胃癌细胞发生内质网应激时的表达
Expression of cytokine induced apoptosis inhibitor 1 in endoplastic reticulum stress in gastric cancer
【摘要】 目的:观察胃癌细胞系BGC823和SGC7901细胞发生内质网应激时CIAPIN1的表达水平。方法:5μg/ml的衣霉素作用细胞6、12和24 h后,采用CCK-8和流式细胞术检测2组细胞的生长抑制;5μg/ml的衣霉素作用24 h后,TUNEL法检测2组细胞的凋亡情况,RT-PCR和免疫印迹检测葡萄糖调节蛋白78(GRP78)和CIAPIN1的表达水平。结果:衣霉素可以诱导2组细胞发生内质网应激及其介导的细胞凋亡;GRP78的表达显著升高,同时CIAPIN1的表达显著降低,且在BGC823和SGC7901细胞间差异均没有统计学意义。结论:CIAPIN1的表达在胃癌细胞发生内质网应激时降低,可能在胃癌的发生及增殖凋亡过程中起到重要作用。
【Abstract】 Objective:To investigate the expression level of cytokine induced apoptosis inhibitor 1(CIAPIN1) induced by endopkstic reticulum stress in gastric cancer cell line BGC823 and SGC7901,and explore the relationship of CIAPIN1 and endoplastic reticulum stress-induced apoptosis in gastric cancer.Methods:CCK-8 assay was used to evaluate the effects of tunicamycin treatment on growth inhibition after 5 μg/ml of tunicamycin treatment for 6,12,24 h;Apoptosis were tested by flow cytometry and TUNEL method,and RT-PCR and Western blotting was performed to detect the expression of GRP78 and CIAPIN1 in the two groups treated with tunicamycin at 5 μg/ml for 24 h.Results:5 μg/ml of tunicamycin treatment for24 h could obviously inhibit proliferation and induce apoptosis in the two groups(P<0.01);The expression of GRP78 was statistically higher.CIAPIN1 was statistically lower simultaneously in the two groups treated by tunicamycin than that in the control group,but there was no statistical difference between BGC823 and SGC7901.Conclusion:The expression of CIAPIN1 is down-regulated in endoplastic reticulum stress-induced apoptosis in gastric cancer cell line BGC823 and SGC7901,and may play an important role in the proliferation and apoptosis of gastric cancer.
