氢水对脑缺血再灌注大鼠脑组织一氧化氮及其合酶的影响
Influence of hydrogen on nitric oxide content and nitric oxide synthase activity in rat tissues following focal cerebral ischemia/reperfusion injury
【摘要】 目的:探讨氢水治疗对急性脑缺血/再灌注损伤引起的脑组织一氧化氮(NO)含量和一氧化氮合酶活力改变的影响。方法:成年SD大鼠随机分为假手术组、模型组、氢水治疗组。采用线栓法制作大鼠大脑中动脉缺血再灌注(MCAO)模型,免疫组织化学测定大脑皮层一氧化氮合酶的表达,硝酸还原酶法测定缺血侧脑组织匀浆中NO含量,分光光度计法测定脑组织提取液中一氧化氮合酶的活性。结果:术后6 h时,模型组脑组织一氧化氮合酶阳性细胞灰度值、NO浓度及一氧化氮合酶活性均较假手术组显著升高,而氢水治疗后其水平较模型组显著下降,但仍高于假手术组。治疗24 h时,模型组一氧化氮合酶阳性细胞灰度值、NO浓度及一氧化氮合酶活性仍显著高于假手术组,但治疗组与模型组相比差异无统计学意义。结论:氢水治疗可通过早期减少缺血后脑组织中NO含量及降低一氧化氮合酶的活性来减轻脑缺血/再灌注损伤。
【Abstract】 Objective:To study the role and mechanism of hydrogen on acute cerebral ischemic/reperfusion injury.Methods:Adult rats were divided into a sham operation group,a model group and a hydrogen therapy group.The middle cerebral artery occlusion(MCAO) model induced by inserting a monofilament suture was adopted.The content of NO,and the activity of NOS in brain homogenate was determined.Results:The expression of NOS,the content of NO and the activity of NOS in the injured cerebral brain was apparently increased in the model group in 6 h after operation compared with the sham operation group;but all of those decreased after hydrogen therapy compared with the model group.Similarly,the expression of NOS,the content of NO and the activity of NOS in the injured cerebral brain was apparently increased in the model group in 24 h after operation compared with the sham operation group;but all of those had no significant changes after hydrogen therapy compared with the model group.Conclusion:Hydrogen has a protective effect on cerebral ischemia at the early stage and the possible mechanism may be related to the reduction in the content of NO and the activity of NOS.
