Toll样受体4多态性调节白细胞介素1-α表达与宫颈癌相关性

【摘要】 目的:研究Toll样受体4(TLR4)多态性与宫颈癌相关性及可能的机制。方法:246名宫颈癌患者和年龄相匹配的246名健康对照个体TLR4 A896G、D299G和T399I多态性位点基因型用聚合酶链反应-限制性片段长度多态性(PCRRFLP)检测;ELISA方法检测宫颈癌组织匀浆中白细胞介素1-α(IL-1α)、白细胞介素-8(IL-8)、转化生长因子-β(TGF-β)和肿瘤坏死因子-α(TNF-α)蛋白水平。结果:与T399I CC基因型相比,CT合并TT基因型显著增加宫颈癌的患病风险,而携带T399IT等位基因的个体患病风险是C等位基因个体的1.912倍,TLR4 A896G和D299G多态性基因型在病例-对照组中频率无差异;IL-1α浓度在T399I CT合并TT基因型个体宫颈癌组织中的表达高于T399I CC组个体。结论:TLR4T399I CT合并TT基因型可增加个体患宫颈癌的风险,其机制可能是调节宫颈癌组织中IL-1α蛋白表达。

【Abstract】 Objective:To investigate the association of TLR4 D299 G,T399I and A896 G polymorphisms and cervical carcinoma,and to explore the effects of polymorphisms on cytokine in the cervical carcinoma biopsies.Methods:The genotypes of these three loci among 246 patients with cervical carcinoma and 246 healthy controls were determined by polymerase chain reaction-restriction fragment lengthy polymorphism(PCR-RFLP).Cytokines in cervical carcinoma biopsies were measured by enzyme-linked immunosorbent assay(ELISA).Results:It showed that CT combined TT genotype of T399 I was associated with increased cervical carcinoma risk at OR=1.985(95%CI:1.286-3.135,P<0.01),individuals with T allele of T399 I showed a 1.912-fold increase in cervical carcinoma risk as compared with the T399 I C allele(95%CI:1.315-2.817,P<0.01).Protein levels of IL-1α was significantly elevated in patients with T399 I CT and TT genotypes.Conclusion:Our data suggest that CT combined TT genotype of TLR4 T399 I is associated with increasing cervical carcinoma risk,and the possible mechanism may be modified IL-lα protein expression in the biopsies of cervical carcinoma.
