定量蛋白质组学技术在阿尔茨海默症临床标志物遴选中的研究进展
The research progress of quantitative proteomics in clinical biomarker discovery in Alzheimer’s disease
【摘要】 阿尔茨海默症(Alzheimer’s disease,AD)是一种以行为功能紊乱、神经元逐渐丢失为主要特征的退行性疾病,它对人类的生活质量和生命造成了极大威胁.随着临床标志物(clinical biomarker)概念的提出和定量蛋白质组学的研究进展,对于促进AD病诊断、监测疾病进程以及评估现有治疗手段等方面提出了新的思路,但临床标志物的遴选存在许多技术难点.定量蛋白质组学利用其能够快速分析和准确鉴定疾病中功能蛋白的特点,大大提高了遴选临床标志物的效率.本文综述了定量蛋白质组学技术在遴选AD中临床标志物的应用进展.
【Abstract】 Alzheimer’s disease(AD) is a neurodegenerative disease characterized with behavioral disturbances and neuronal loss, which threaten people’s life and lower their life quality. In recent years, the proposal of concept "clinical biomarker" provide new ideas for promoting AD diagnosis, monitoring disease progress and assessing existed treatment. However, there are many technical difficulties in discovering clinical biomarker in AD. With its ability about capture functional protein precisely, quantitative proteomics plays an important role in improving the efficiency of clinical biomarker discovery. In this review, we summarize the application of quantitative proteomics in discovering clinical biomarker in AD.
