【摘要】 目的观察肿瘤坏死因子-α(TNF-α)对体外培养的人肾小管上皮细胞(HK-2)增殖及表达结缔组织生长因子(CTGF)蛋白的影响。方法采用DMEM/F12培养基体外培养HK-2细胞,分别用不同浓度的TNF-α刺激HK-2细胞,用噻唑蓝(MTT)法检测HK-2细胞的增殖,用流式细胞术(FCM)检测细胞周期,ELISA方法检测培养细胞的上清液中CTGF蛋白含量。结果与对照组(不加TNF-α)相比,0.1ng/mLTNF-α组、0.5ng/mL TNF-α组、1.0ng/mL TNF-α组、5ng/mL TNF-α及10ng/mL TNF-α组,MTT法测定的吸收光度A值、细胞增殖指数、ELISA方法检测培养细胞的上清液中CTGF蛋白含量,均有极显著性升高(P<0.01),并随着TNF-α浓度的增加而递增(P<0.05或P<0.01)。但是,当培养液中TNF-α浓度到达1.0ng/mL后达到高峰,即在1.0ng/mL TNF-α、5ng/mL TNF-α及10ng/mL TNF-α组间,上述指标均无显著性差异(P>0.05)。结论炎症因子TNF-α能促进肾小管上皮细胞的增殖、分泌致纤维化因子CTGF,从而在慢性肾脏疾病的发生发展中发挥重要作用。更多还原

【Abstract】 Objective To investigate the effect of tumor necrosis factor-alpha( TNF-α) on cell proliferation of HK-2 and protein expression of connective tissue growth factor( CTGF) by HK-2. Method HK-2 cells were cultured in vitro and treated with TNF-α by Respective Concentration of 0ng / mL,0. 1ng / mL,0. 5ng / mL,1. 0ng / mL,5ng / mL and 10ng / mL. The proliferation of HK-2 cells were assessed by MTT. Cell cycle was detected by flow cytometry technology. CTGF protein was assessed by enzyme linked immunosorbent assay(ELISA) in the supernatant. Results The absorption value A,Cell proliferation index and CTGF Protein content in different densities of TNF-α were higher significantly than those in control group(0 ng / mL TNF-α)( P < 0. 05 or P < 0. 01). Those items were higher with TNF-α concentrations increased. But Those indicators didn,t rise continually when TNF-αdensity was above 1. 0ng / mL. Conclusion Inflammatory factor TNF-α canpromote the cell proliferation and the expression of CTGF in HK-2 cells. This may be one of mechanisms in chronic kidney disease development.更多还原
