钯催化C-N偶联构筑吲哚衍生物反应研究
Study on Palladium Catalyzed C-N Coupling to Construct Indole Derivatives
【摘要】 以炔基苯胺衍生物为底物,通过钯催化C-N偶联合成一类系列吲哚衍生物。优化实验表明,以2%醋酸钯为催化剂,4%三苯基磷为配体,在有机碱的DMF溶液中加热至130℃为优化实验条件。产物结构经IR、1H NMR、13C NMR、HRMS分析确证,其中化合物2e经X射线单晶衍射分析确证。
【Abstract】 A series of indole derivatives were synthesized from 2-ethynylaniline derivatives by palladium catalyzed C-N coupling. The following reaction conditions had been selected to conduct these studies: substrate( 1equiv) in the presence of 2( mol) % of palladium( II) catalyst and 4( mol) % of Ph3 P with n-Bu3N( 2 equiv) as an additive in DMF at 130℃. The products were characterized by IR,1H NMR,13 C NMR and HRMS diffraction analysis,and the structure of compound 2e was confirmed using single-crystal X-ray analysis.
