高血压脑出血急性期肝阳化风证与恢复期阴虚风动证的蛋白质组学比较研究

【摘要】 目的动态观察高血压脑出血患者急性期肝阳化风证(简称肝阳化风证)、恢复期阴虚风动证(简称阴虚风动证)外周血单个核细胞的蛋白质差异表达变化,分析脑出血不同发病阶段不同中医证型的蛋白质表达规律,探讨脑出血肝阳化风证、阴虚风动证的本质内涵。方法双向凝胶电泳(2-DE)分离脑出血急性期肝阳化风证(发病3 d)、恢复期阴虚风动证(发病3月)、健康对照组(简称对照组)外周血单个核细胞蛋白质,采用考马斯亮蓝染色,PDQuest软件对三组2-DE图谱进行差异分析,选取差异蛋白质点进行MALDI-TOF-MS质谱分析,Mascot软件搜索数据库初步鉴定蛋白质。结果获得了重复性较好的高血压脑出血肝阳化风证、阴虚风动证2-DE图谱;质谱初步鉴定出3个差异表达蛋白,分别为肌动蛋白、假想蛋白、纤维蛋白原α链前体,其中肌动蛋白在脑出血肝阳化风证组、阴虚风动证均较对照组表达下调,而阴虚风动证组又较肝阳化风证组表达下调;纤维蛋白原α链前体在脑出血肝阳化风证组表达上调,而阴虚风动组表达下调。结论高血压脑出血急性期肝阳化风证与恢复期阴虚风动证存在差异蛋白的表达,其中肌动蛋白与纤维蛋白原α链前体可能与脑出血肝阳化风证、阴虚风证动的发展密切相关。。

【Abstract】 Objective This paper is to investigate the dynamic changes of peripheral blood mononuclear cell proteins expression on the acute liver yang into wind, and wind formation from Yin deficiency syndrome of hypertensive intracerebral hemorrhage(HCH) patients. The protein expression pattern of different TCM syndromes in different stages was analyzed, and then it can explore the essence of the two TCM syndromes from proteomics. Methods Immobilized pH gradient two-dimensional polyacrylamide gel electrophoresis(2-DE) was applied to separate the PBMCs proteins of HCH patients with acute liver yang into wind(3 days of illness) and wind formation from Yin deficiency syndrome during recovery period(3 mouths of illness) and healthy control group(control group). Kaumas Coomassie brilliant blue staining was used. PDQuest software was applied to analysis the difference of three groups of 2-DE fingerprinting differential protein spots and select these spots. The differential protein spots were obtained the peptide mass fingerprints with Matrix-assisted laser desorption/ionization-time of flight mass spectrometry(MALDI-TOF-MS), and then some of differential proteins were identified with Mascot search software. Results A good 2-DE pattern with high reproducibility was obtained. Three differentially expressed proteins: actin, hypothetical protein and fibrinogen alpha chain precursor were identified primarily by MS. The expression of actin wind formation from in Yin deficiency syndrome group and acute liver yang into wind syndrome group was down-regulated, while the expression in wind formation from Yin deficiency group was lower than acute liver yang into wind group. The expression of fibrinogen alpha chain precursor in the liver yang into wind syndrome group was up-regulated, while the wind formation from Yin deficiency group down-regulated. Conclusion Expression proteins are differential between hypertensive cerebral hemorrhage patients with acute liver yang into wind and wind formation from Yin deficiency syndromes during recovery period.Expressions of actin and fibrinogen alpha chain precursor may be closely related to the development of wind formation from Yin deficiency, and liver yang into wind syndromes.
