血管紧张素Ⅱ和白介素6在静脉留置针致血管损伤中的作用

【摘要】 目的探讨血管紧张素Ⅱ(AngⅡ)和白介素6(IL-6)在静脉留置针致血管损伤中的作用。方法选新西兰大耳兔40只,随机分为4组,每组10只,其中一组作为对照组,常规饲养1 W后留取静脉血和双耳外缘中段静脉血管组织。其余3组为实验组,兔双耳外缘静脉留置针穿刺置管,注射生理盐水,20 mL/次,1次/d。3组分别留置3 d、5 d、7 d,达到留置时间后抽取静脉血,麻醉后留取局部血管组织。用放射免疫法检测血液及血管组织中的AngⅡ和IL-6的含量。结果血液检测:AngⅡ含量,5d组、7d组明显高于对照组(P<0.01);实验组组间比较差异有统计学意义(P<0.01,P<0.05),随着留置时间的延长AngⅡ依次增高;IL-6含量,7 d组明显高于对照组、3 d组和5 d组(P<0.01)。组织检测:AngⅡ含量,3组实验组与对照组比较差异有统计学意义(P<0.01),实验组组间比较,5 d组、7 d组高于3 d组(P<0.05);IL-6含量,5 d组、7 d组明显高于对照组(P<0.01);实验组组间比较差异有统计学意义(P<0.01)。结论静脉留置针刺激可使AngⅡ和IL-6含量增高,引起血管收缩痉挛,促使血管炎症反应发生,而致血管损伤。

【Abstract】 Objective To study the role of angiotensinⅡ and Interleukin-6 in vascular injury induced by venous indwelling needle. Methods Totally 40 New Zealand rabbits were divided into four groups randomly with 10 rabbits each. One group without any measures as a control group , after one week feeding specimens venous blood and the middle of venous vascular tissue on the ears outer edge. Two edged-veins in both ears of other three groups were set intravenous indwelling needle as experimental group, normal saline 20ml was infused via the catheter every day. Specimens venous blood and vascular tissue with anesthesia after indwelling 3 days, 5 days and 7 days. The contents of AngⅡand IL-6 in blood and vascular tissue were measured by radioimmunoassay. Results The contents of AngⅡ in plasma: 5 days group and seven days were significantly higher than that of the control group (P<0.01); the three groups was compared has differences (P<0.01,P<0.05), and the contents of AngⅡ in turn increased with the the indwelling time extension.The contents of IL-6 in serum:seven days group were significantly higher than that of the control group,, 3 and 5 days groups(P<0.01). The contents of AngⅡ in vascular tissue: they are significant differences in the whole experimental groups and control group (P<0.01); compared with the three experimental groups, 5 days and 7 days groups are higher than 3 days group (P<0.05). The contents of IL-6 in vascular tissue: 5 days group and 7 days were significantly higher than that of the control group (P<0.01); and they are significant differences in the three experimental groups by compared (P<0.01). Conclusion The contents of AngⅡand IL-6 will be increased by set intravenous indwelling needle, it caused vasoconstriction and spasm to injure vascular.
