eIF3a和HE4表达与卵巢癌的相关性

【摘要】 目的:探讨真核细胞翻译起始因子3a(eukaryotic translation initiation factor 3,subunit A,e IF3a)和人附睾蛋白4(human epididymis protein 4,HE4)与卵巢癌发生、发展的关系及e IF3a和HE4表达之间的相关性。方法:分别采用RTPCR和免疫组织化学法检测181例卵巢癌患者、卵巢良性肿瘤患者及正常卵巢组织中e IF3a与HE4的m RNA和蛋白表达情况。结果:e IF3a基因和HE4基因在正常卵巢、卵巢良性肿瘤及卵巢癌组织中的m RNA及蛋白表达水平均差异有统计学意义(P<0.05)。e IF3a基因和HE4基因m RNA表达在卵巢癌组织与正常卵巢组织、卵巢良性肿瘤组织与正常卵巢组织中差异均有统计学意义(P<0.05);其中正常卵巢组织e IF3a基因m RNA表达水平高于卵巢良性肿瘤和卵巢癌组织,而HE4基因m RNA表达水平从正常卵巢、卵巢良性肿瘤到卵巢癌组织中逐渐升高;HE4基因m RNA表达水平卵巢癌高于卵巢良性肿瘤组织(P<0.001)。e IF3a蛋白在正常卵巢、卵巢良性肿瘤及卵巢癌组织中的阳性表达率分别为0,66.7%和81.0%,HE4蛋白在正常卵巢组织、卵巢良性肿瘤组织及卵巢癌组织中的阳性表达率分别为0,27.8%和56.2%。e IF3a蛋白和HE4蛋白表达阳性率在卵巢癌组织与卵巢良性肿瘤组织、卵巢癌组织与正常卵巢组织、良性肿瘤组织与正常卵巢组织中差异均有统计学意义(P<0.001)。e IF3a蛋白和HE4蛋白表达水平呈正相关(r=0.35,P<0.05)。结论:e IF3a和HE4的表达与卵巢癌发生相关,二者可能通过细胞外调节蛋白激酶通路相互联系。

【Abstract】 Objective: To investigate the correlation between eukaryotic translation initiation factor 3, subunit A(e IF3a) and human epididymis protein 4(HE4) expression and ovarian cancer. Methods: RT-PCR or immunohistochemistry was used to examine e IF3 a and HE4 m RNA or protein expression in ovarian tissues from patients with ovarian cancer(n=181) or benign ovarian tumors, or from the healthy women. Results: There were significant differences in m RNA and protein expression of e IF3 a and HE4 among normal ovarian tissues, benign ovarian tumor tissues, and ovarian cancer tissues(P<0.05). There were significant differences in m RNA expression of e IF3 a and HE4 between the normal tissues and the ovarian cancer tissues, or between the benign ovarian tumor tissues and the normal tissues(P<0.001). The m RNA expression of e IF3 a in the normal ovarian tissues was significantly higher than that in the benign ovarian tumor tissues or that in the ovarian cancer tissues. The m RNA expression of HE4 was gradually increased from the normal ovarian tissues, the benign ovarian tumor tissues to the ovarian cancer tissues. The m RNA expression of HE4 in the ovarian cancer tissues was significantly higher than that in the benign ovarian tumor tissues(P<0.001). Positive expression rates for e IF3 a or HE4 protein in normal, benign tumor, and cancer tissues were 0, 66.7%, and 81.0% or 0, 27.8%, and 56.2%, respectively. There were significant differences in positive expression rates of e IF3 a protein and HE4 protein between the ovarian tumor tissues and benign ovarian tumor tissues, between the ovarian cancer tissues and the normal ovarian tissues, or between the benign ovarian tumor tissues and the normal ovarian tissues(P<0.001). The e IF3 a protein expression was positively correlated with HE4 protein expression(r=0.575, P<0.05). Conclusion: The expressions of e IF3 a and HE4 are associated with ovarian cancer, and extracellular regulated protein kinases may play a role in the interaction between eIF3 a and HE4.
