埃克替尼对ACC-M细胞凋亡及Bcl-2,Bax蛋白表达的影响
Effect of Icotinib on the Apoptosis of ACC-M Cells and the Protein Expression of Bcl-2 and Bax
【摘要】 按照0(对照组)、10、20、40μmol·L-1浓度配制盐酸埃克替尼溶液,分别作用于人涎腺腺样囊性癌细胞株(ACC-M)24、48、72h,用流式细胞仪测药物作用后ACC-M细胞凋亡率;同时利用免疫细胞化学方法和间接免疫荧光联合流式细胞技术,检测它在体外对ACC-M细胞Bcl-2、Bax蛋白表达的影响.结果显示,细胞凋亡率随盐酸埃克替尼浓度与作用时间的增加而增加,经统计学分析,细胞凋亡率与盐酸埃克替尼浓度、作用时间成正相关,药物作用于ACC-M细胞后Bcl-2蛋白表达降低,Bax蛋白表达增加.实验表明,盐酸埃克替尼可能通过降低Bcl-2蛋白的表达,增加Bax蛋白的表达,诱导ACC-M细胞凋亡.
【Abstract】 In this study,we used different concentrations of Icotinib hydrochloride[respectively 0(for control group),10,20and 40μmol·L-1]to act on ACC-M cells of human beings for respectively 24,48,and 72h.And then we tested the apoptosis rate with flow cytometer.In the meantime,we used the techniques of immunocytochemistry and indirect immunofluorescence to test the effect of Icotinib upon the protein expression of Bcl-2and Bax in ACC-M cells in vitro.The tests showed that the apoptosis rate increased with the enhancement of the consistency of Icotinib hydrochloride and the amount of time used.Statistical analysis showed that there was a positive correlation between the apoptosis rate and the consistency of Icotinib hydrochloride and the amount of time used in the process,and that the protein expression of Bcl-2decreased while that of Bax increased under the influence of Icotinib.Our experiment shows that Icotinib hydrochloride may induce the apoptosis of ACC-M cells via reducing the expression of Bcl-2and increasing that of Bax.
