辛酸钠对奶牛乳腺上皮细胞脂肪酸合成相关酶表达的影响

【摘要】 为研究辛酸钠对奶牛乳腺上皮细胞脂肪酸合成关键酶乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FAS)和硬脂酰辅酶A去饱和酶(SCD)表达的影响,采用0.5、1.0、2.0mmol/L的辛酸钠处理体外培养的奶牛乳腺上皮细胞,采用实时荧光定量PCR(qRT-PCR)检测ACC、FAS、SCD基因相对表达丰度,Western blot检测ACC、FAS、SCD蛋白相对表达水平。结果显示:与不添加辛酸钠组相比,辛酸钠以浓度依赖的方式显著抑制ACC、FAS、SCD的表达。辛酸钠能抑制奶牛乳腺上皮细胞脂肪酸合成关键酶和硬脂酰辅酶A去饱和酶的表达。

【Abstract】 To investigate the effects of sodium octanoate on the expression level of key fatty acid synthesis enzymes ACC,FAS and SCD in desaturation of dairy mammary epithelial cells(DCMECs) cultured in vitro.In the experiment,the mRNA expression levels of ACC,FASand SCDwere determined by qRT-PCR and the protein expression levels of ACC,FAS and SCD were determined by Western blot. The results showed as follows:compared with the group without sodium atanoate,the gene expression and protein expression of ACC,FAS and SCD were inhibited significantly by in sodium octanoate in concentration-dependent manner from 0.5 mmol/L to 2 mmol/L.This study indicated that sodium octanoate could inhibit the gene and protein expression of key fatty acid synthesis enzymes and stearyl coenzyme A desaturation enzyme in dairy mammary epithelial cells.
