【摘要】 目的通过体外药敏试验,检测目前常用化疗药物对体外培养的非小细胞肺癌细胞的敏感性,为临床对化疗药物的选择提供参考,从而减少临床对化疗药物选择的盲目性,更好地实行个体化治疗。方法通过改良四氮唑盐(tetrazolium salt,MTT)比色法进行肺癌细胞体外药敏试验,对41例肺癌患者的新鲜肿瘤标本进行体外培养和化疗药物敏感性检测,并对其结果进行比较。结果 PTX、NVB、DDP、L-OHP的敏感率均在40%以上,提示非小细胞肺癌对以上药物敏感,而CPT-11、VCR、THP、ADM、EPI、IFO、CBP的敏感率均低于30%,提示在非小细胞肺癌化疗过程中应用以上药物可能存在耐药,非小细胞肺癌中腺癌和鳞癌对16种化疗药物的敏感性也有一定的差异性。结论术后肺腺癌的辅助化疗中应首选PTX、5-FU、DDP、L-OHP、CPT、VP-16、Docetaxel;肺鳞癌的辅助化疗中应首选PTX、DDP、L-OHP、NVB,VCR、THP、ADM均100%耐药,临床应用中应注意避免应用这些药物。更多还原

【Abstract】 Objective Through in vitro drug susceptibility testing,to detect sensitivity of chemotherapy drugs on cultured non-small cell lung cancer cells,provide reference for clinical choice of chemotherapy drugs and help to choose the clinical chemotherapy drgus and better individual treatment. Methods In vitro susceptibility in the lung cancer cells was tested through improved tetrazolium salt( MTT) assay. A total of 41 fresh tumor samples of lung cancer patients were cultured in vitro and drug susceptibility testing of chemotherapy was performed. The results were compared. Results The sensitivities of PTX,NVB,DDP,L-OHP were above 40 %,suggesting that non-small cells with lung cancer were sensitive to these drugs. The sensitivities of CPT-11,VCR,THP,ADM,EPI,IFO,CBP were less than 30 %,suggesting that non-small cells with lung cancer had drug resistance to these drugs. The sensitivities of nonsmall cell lung adenocarcinoma and squamous cell were different to 16 kinds of chemotherapy drugs. Conclusion PTX, 5-FU,DDP,L-OHP,CPT,VP-16,Docetaxel are preferred for postoperative adjuvant chemotherapy in lung cancer. PTX,DDP,L-OHP,NVB are preferred for lung squamous cell carcinoma of adjuvant chemotherapy. Patients are 100 percent resistant to VCR,THP,ADM,which should be avoided in clinic.更多还原
