氯化消毒饮用水有机提取物对大鼠肝脏芳香烃受体和相关基因表达的影响

【摘要】 目的探讨氯化消毒饮用水水样中有机提取物染毒对大鼠肝脏芳香烃受体(aryl hydrcarbon receptor,AhR)和相关基因表达的影响。方法于2012年7—9月(丰水期)采集贵阳市某地水厂的管网末梢水水样并制备有机提取物。将40只清洁级SD大鼠随机分为4组,分别为对照(玉米油)组和5、20、80 L/kg饮用水有机提取物染毒组,每组10只,雌雄各半。采用经口灌胃方式进行染毒,每天1次,连续染毒12周。采用实时荧光定量PCR法检测肝脏中Ah R、热休克蛋白(HSP90)、芳香烃受体核转位蛋白(ARNT)和细胞色素1A2(CYP1A2)mRNA的表达水平,采用Western Blot法检测肝脏中AhR、HSP90、ARNT与CYP1A2蛋白的表达水平。结果与对照组比较,5、20、80 L/kg饮用水有机提取物染毒组大鼠肝脏中HSP90的mRNA和蛋白表达水平均增高,20、80 L/kg组大鼠肝脏中AhR、ARNT与CYP1A2 mRNA和蛋白的表达水平均增高,差异有统计学意义(P<0.05)。结论在本实验条件下,氯化消毒饮用水有机提取物暴露可活化AhR通路。

【Abstract】 Objective To know the expression of Ah R and related genes in rats exposed to organic extracts from chlorinated drinking water. Methods The organic compounds were extracted from the pipeline water in Guiyang from July to September(wet season) in 2012. A total of 40 SD rats were randomly divided into solvent control group(with corn oil) and three exposure groups with 10 rats in each group(half male and female). The exposure groups were administered respectively with organic extracts with doses of 5,20,80 L/kg body,once a day, through gavage. The exposure lasted for 12 weeks. Real-time quantitative PCR was used to measure m RNA expression level of Ah R,HSP90,ARNT and CYP1A2 and Western blot was used to detect protein expression level of Ah R,HSP90,ARNT and CYP1A2. Results Compared with the control group,m RNA and protein expression levels of HSP90 in 5,20 and 80 L/kg groups were up-regulated(P <0.05),while m RNA and protein expression levels of Ah R,ARNT and CYP1A2 were obviously down-regulated only in 20,80 L/kg groups(P<0.05). Conclusion Under the condition of this experiment,exposing to organic extracts from chlorinated drinking water can activate the Ah R pathway.
