苹果酵素对微囊藻的溶藻特征研究
Research on algicidal charactistic of apple enzyme for lysis of Microcystis
【摘要】 目的研究自制苹果酵素对微囊藻的溶解作用,并探讨其溶藻机制。方法在微囊藻藻液中加入不同体积(0~3 ml)、不同稀释度(0~10-5倍)及经不同处理[原液、经0.22μm滤膜抽滤2次、高温灭菌(121℃、0.1 MPa灭菌20 min)、以12 000 r/min离心(离心半径3 cm)15 min、0.25%的胰蛋白酶处理15 min]的苹果酵素进行溶藻实验,测定叶绿素a的含量并计算清除率。结果随着加入苹果酵素量的增加,叶绿素a的清除率先升高后下降,在加入0.6 ml时除藻作用最强,第4天的清除率达到最大,为94.48%;随稀释倍数的增加,叶绿素a的清除率先升高后下降,稀释10-1倍时的清除率最强,在第4天达最大,为95.27%;经过不同处理的苹果酵素都能产生溶藻作用,第4天原液、经0.22μm滤膜抽滤2次、高温灭菌、以12 000 r/min离心(离心半径3 cm)15 min、0.25%的胰蛋白酶处理15 min的叶绿素a清除率分别为81.40%,84.36%,80.18%,79.52%和79.30%。结论该苹果酵素具有溶解微囊藻的作用,其溶藻机制为产生一种耐高温的非蛋白类物质间接作用溶藻。
【Abstract】 Objective To study the algicidal effect of homemade apple enzyme to Microcystis,and discuss the algicidal mechanism. Methods The content of chlorophyll-a and the clearance rate were determined on adding apple enzyme with the volume from 0 to 3 ml, the dilution of 0 to 10-5times, and different processing [the stoste, the stoste needs to be collated twice through the filter of 0.22 μm, high-temperature sterilization(121 ℃,0.1 MPa,20 minutes), centrifugation of 15 minutes with 12 000 r/min(r =3 cm), and 0.25% trypsin treatment for 15 minutes in Microcystis]. Results With the increasing of apple enzyme,the clearance of chlorophyll-a increased and then decreased,and the function was maximum when added 0.6 ml apple enzyme into the fluid of Microcystis, the clearance of chlorophyll-a reached 94.475% at the fourth day. The apple enzyme was diluted with distilled water, and the clearance of chlorophyll-a increased and then decreased with the increasing dilution multiple,when it was diluted by 10-1times, the clearance of chlorophyll-a reached 95.27% at the fourth day. The clearance of these different treatments at the fourth day was as follows: 81.40% for stoste, 84.36% for twice filtration by 0.22 μm filter, 80.18% for high-temperature sterilization(121 ℃,0.1 MPa,20 minutes), 79.52% for 15-minute centrifugation with 12 000 r/min(r=3 cm), 79.30% for 15- minute treatment with 0.25% trypsin. Conclusion The apple enzyme used in the study has effect on algicidal Microcystis, the algicidal mechanism is indirect action by producing a high temperature resistance of nonprotein substance.
