氧化型低密度脂蛋白提高小鼠骨髓间充质干细胞粘附力

【摘要】 分别采用5、10和20μg/mL的氧化型低密度脂蛋白(oxidized low-density lipoprotein,ox-LDL)处理小鼠骨髓间充质干细胞(bmMSCs)6h,通过CellTracker预染小鼠单核细胞,检测单核与bmMSCs之间的粘附率,研究ox-LDL对bmMSCs粘附的影响.利用Western blot检测粘附分子VCAM-1和ICAM-1于ox-LDL处理后在bmMSCs内的表达情况.结果显示:bmMSCs阳性表达间充质干细胞特异性标记分子CD44和CD90;ox-LDL可显著提高bmMSC与单核细胞之间的粘附(P<0.05),增强粘附分子VCAM-1和ICAM-1的表达(P<0.05),且具有剂量效应.以上结果表明,ox-LDL通过增强粘附分子VCAM-1和ICAM-1的表达进而增强bmMSCs对单核细胞的粘附,其或许可作为一种提高间充质干细胞粘附和提高干细胞移植后存活率的诱导剂.

【Abstract】 To study the effect of oxidized low-density lipoprotein(ox-LDL)on adhesion of mouse bone marrow mesenchymal stem cells(bmMSCs),BmMSCs were treated with 5,10 and 20μg/mL ox-LDL for 6h.Adhesive rate of mouse monocytes on bmMSCs was measured through pre-dye of monocytes with CellTracker.Expression of adhesive molecules VCAM-1and ICAM-1in bmMSCs were measured by Western blot assay after treatments with ox-LDL.BmMSCs were positive for MSC specific makers CD44 and CD90.Treatments with ox-LDL markedly increased adhesive rate of monocytes on bmMSCs,and enhanced expression of VCAM-1and ICAM-1in a dose-dependent manner(P<0.05).These results indicate that ox-LDL can enhance adhesion of bmMSCs to monocytes via increase of expression of adhesion molecules VCAM-1and ICAM-1,and maybe serve as an inducer to increase adhesion and survivability of MSCs after transplant.
