SRp20对口腔癌细胞丙酮酸激酶前体mRNA可变剪切的调控作用
SRp20 Regulates the Alernative Splicing of Pyruvate Kinase Pre-mRNA in Oral Squamous Carcinoma Cells
【摘要】 目的:研究SRp20参与口腔癌细胞中丙酮酸激酶前体mRNA可变剪切的调控机制。方法:采用siRNA在口腔癌细胞Cal27和Tca8113中沉默SRp20的表达,再以RT-PCR检测丙酮酸激酶的M1和M2亚型的表达水平。结果:SRp20的特异性siRNA转染Cal27和Tca8113细胞,显著降低了SRp20的表达水平。在Cal27细胞和Tca8113细胞中,抑制SRp20的表达后,M2的表达水平下降,M1与M2表达水平的相对比例显著增加。结论:在口腔癌细胞中,SRp20调控丙酮酸激酶前体mRNA可变剪切,利于M2亚型丙酮酸激酶的表达。
【Abstract】 Objective:To investigate the function of SRp20in the alternative splicing of pyruvate kinase in oral squamous carcinoma cells.Methods:The expression of SRp20was suppressed by siRNA(small interfering RNA)in Cal27or Tca8113cells.The expression levels of pyruvate kinase M1 and M2 were analyzed by RT-PCR.Results:SRp20siRNA efficiently down-regulated SRp20expression in Cal27and Tca8113cells.Knockdown of SRp20decreased M2 and increased the relative ratio of M1 /M2.Conclusion:SRp20regulates the alternative splicing of pyruvate kinase premRNA and increases the expression of pyruvate kinase M2 in oral squamous carcinoma cells.
