菠菜育种材料的遗传多样性分析

【摘要】 利用形态学分类法和AFLP分子标记技术对45份菠菜(Spinacia oleracea L.)育种材料的遗传多样性和亲缘关系进行了研究。结果表明,从256对引物中筛选出18对扩增效果较好的引物,共扩增到960个条带,146个多态性位点,PIC值(平均多态信息量)为15.2%。AFLP标记技术的聚类分析表明,第一类群主要以欧美资源和有欧美血统的杂交种为主;第二类群主要以日本资源和含有日本血统的杂交种为主;第三类群以中国本土品种为主。从聚类结果看,与形态学的分类结果基本一致,而且聚类结果与地域来源和形态特征有一定的对应关系。

【Abstract】 The genetic diversity and phylogenetic relationship of 45(Spinacia oleracea L.) breeding materials of spinach were studied by traditional taxonomy and AFLP markers.The results showed that eighteen polymorphic primes pairs were screened from 256 primer pairs and 146 polymorphic bands were observed from total 960 amplified bands.The value of PIC was15.2%.The genetic diversity of 45 spinach breeding materials with AFLP molecular markers was clustered into three groups.The first group was mainly the Europe and America varieties.The second group was mostly the Japan varieties.The third group was the Chinese varieties.The results were consisted with the traditional classification.The clusters were associated with the origins and the morphological characters of the breeding materials tested.
