低次烟叶中茄尼醇的超声波辅助提取与纯化

【摘要】 采用超声波辅助溶剂法对低次烟叶中茄尼醇进行提取并通过硅胶柱层析和重结晶对其进行纯化。由正交试验得出超声波辅助石油醚提取低次烟叶中茄尼醇的最佳工艺条件为温度50℃,时间60 min,料液比1∶15(m/V,g∶mL),超声波功率200 W,在此条件下,茄尼醇提取率达到0.66%。茄尼醇粗提物经过皂化反应含量提高了20%,皂化后产物通过硅胶柱层析进行分离,纯度达到81%,产率0.4%。此纯度的茄尼醇继续用乙醇重结晶精制,纯度可达到90%,收率86%。

【Abstract】 The ultrasonic-assisted solvent extraction of solanesol from inferior tobacco leaf and the purification of solanesol by silica gel column chromatography and crystallization was studied. The results of the rhombic experiments showed that the optimal values of parameters for the ultrasonic-assisted petroleum ether extraction of solanesol from inferior tobacco leaf were extraction temperature 50℃, ultrasonic treatment time 60min, material-solvent ratio 1 ∶15(g ∶mL), ultrasonic power 200W. Under these conditions, the extraction rate of solanesol reached 0.66%. The solanesol content of crude extract increased about 20% by saponification reaction. The saponated solanesol was separated by silica gel column chromatography, with the purity of separated solanesol of 81% and the productivity of 0.4%. The separated solanesol was further purified by recrystallization with ethanol, with the purity of solanesol of 90% and the yield of 86%.
